Study On Quality Analysis And Antioxidant Activities Of Rosa Roxburghii Tratt Fruits And Leaves From Different Planting Bases In Guizhou Province

This research taking three typical Rosa roxburghii Tratt planting bases( Longli County in Qiannan, Qianxi County in Bijie and Qixingguan region in Bijie) in the Karst environment as research object areas,and choose Rosa roxburghii Tratt fruits and leaves as research objects. Choose different points in each base and recorded the small environmental informations. At the same time, collected fruits and leaves samples(total of 64 samples) in each sampling point. The first time apply UV, HPLC, Capacity analysis methods and so on to analysis nutrients of fruits and leaves from different bases respectively. To establish a method for determination of total flavonoids and total polyphenols in Rosa roxburghii Tratt leaves. For the first time by using 5 kinds of antioxidant activity methods in vitro of Rosa roxburghii Tratt fruits and leaves anti-oxidative activities,and analysis foundation and material of fruits and leaves. Combine with the data of soil tested by our research group early stage, and use relevant software to discuss the regular of "chemical composition- antioxidant activity- geographical environment of soil nutrient elements". And make a comprehensive comparison and analysis for the planting environment and the Rosa roxburghii Tratt quality in the three bases. The main results are as follows:(1) The moisture, ash, dietary fiber, tannic acid, titratable acid, total soluble sugar, protein, vitamin C, total flavonoids and total polyphenols contents of Rose roxburghii Tratt in each sampling point from different bases were be tested. Results showed that, Contents in the Rose roxburghii Tratt of the above as follows: 85.06±0.66%, 0.55±0.03g/100 g, 1.85±0.15%, 1.26±0.09%,19.22±0.63%,9.91±0.17%,2.09±0.42g/100 g,2164.25±214.57mg/100 g, 58.02±.41mg/100 g, 2054.15±105.02mg/100 g. From the overall point of view, the quality of Rose roxburghii Tratt fruits of Qianxi County is the best, Longli County is the second, the fruit quality of Qixingguan region in Bijie is not better than the other two.(2)Gallic acid and ascorbic acid as the positive control,with O2-?,?OH,NO2- systems and the total reductive ability and total antioxidant capacity to research antioxidant activities of different sampling points from each planting bases. And evaluate through the EC50 of Rosa roxburghii Tratt on free radical scavenging abilities. The results showed that the ability of free radical scavenging of Rosa roxburghii Tratt fruit is stronger than the same concentration of Gallic acid and Ascorbic acid. The order of the ability of free radical scavenging of Rosa roxburghii Tratt fruit is NO2->?OH> O2-?.The order of antioxidant activity in vitro of fruits is :Qianxi County > Longli county > Qixingguan region.(3) With Gallic acid and Ascorbic acid as the positive control, by using DPPH?, ?OH, NO2- systems and determination of total reducing power and total antioxidant capacity to the different point leaves of Rosa roxburghii Tratt antioxidant activity for the synthetic evaluation of research, and through the EC50 evaluation leaves of Rosa roxburghii Tratt size of all kinds of radical scavenging abilities, investigate different Rosa roxburghii Tratt planting region. Results showing that the leaves of Rosa roxburghii Tratt radical scavenging ability order is DPPH?> · OH > NO2-. NO2- and ·OH scavenging ability order is Qixingguan > Longli > Qianxi, DPPH? scavenging ability and the total reduction and total antioxidant capacity order: Longli > Qixingguan > Qianxi.(4) RDA analysis and Pearson correlation analysis between Components and antioxidant activity of Rosa roxburghii Tratt fruit and microclimate to be used to inspect the effects of Rosa roxburghii Tratt antioxidant activity and environment factor and composition. The study found that the altitude and light is the key environmental factors for components in fruit and the elevation and temperature is the key environmental factors for antioxidant activity in fruit. The Total flavonoids and total polyphenols in fruits are significantly positive correlation with antioxidant activities(P<0.01). The anti-oxidative activities of methods were significantly positive correlation(P<0.01).(5)To study the relationship between the components in the leaves and microclimates and investigate the effect of Antioxidant activity of leaf components. Found that the total flavonoids and polyphenols in the leaf are significant positive correlation(P < 0.01), all them have significantly positive correlation with Antioxidant activity of leaf. There was a positive correlation between different Antioxidant activity methods. Altitude, total nitrogen and Mg in soil are the key environmental factors for compositions of leaf. Light and organic matter in soil are the key environmental factors for Antioxidant activity of leaf. Positively correlated between different antioxidant activity method, in which the total antioxidant capacity and other four methods are significant positive correlation(P<0.01), the total reducing power have no significant correlation between nitrite removal ability, with the other three methods have significant positive correlation(P < 0.05).(6) To establish a method for determination of total flavonoids in and total polyphenols contents in Rosa roxburghii Tratt leaves, and total flavonoids and total polyphenols content of different planting bases were determined.The method validation of the method is stable and reliable, and can be used for determination of total flavonoids and total polyphenol content in Rosa roxburghii Tratt leaves. The total flavonoids and total polyphenol in Rosa roxburghii Tratt leaves contents are: 1512.38 ±973.24mg/100 g, 12546 ± 3460.43mg/100 g.
