This paper aimed to produce an anti-fatigue health wine.The healthy wine used Ginseng,Deer fetus,Rose,Polygonatum sibiricun as the major raw materials,together with white sugar.The optimum extraction process was studied.Percolation method,Hot reflu method,Cold-Merceration were separately adopted to extract.Additionally,effectiveness of these techniques was compared in terms of the total ginsenosides and sensory evaluation.The results showed that compared to the other two extraction techniques,Cold-Merceration was the most effective approach.Then the extraction techniques were optimized in term of the total ginsenosides by orthogonal array design.The orthogonal test was designed based on liquid/material ratio,extraction duration and extraction times.The optimal conditions for Cold-Merceration were as follows: liquid/material ratio 1:10,extraction duration 24 h,2 times.The effects of centrifugal speed and time on the precipitation of the samples were investigated.The analysis showed that the best centrifugation conditions was centrifuging 6 min at a speed of 4000 r/min.Added a certain amount of white sugar or honey to the samples,the values of the sensory evaluation showed that the best amount of the white sugar is 3 g/100 ml.Validity of optimized conditions was verified by UV spectrophotometry.The precision of the total ginsenosides in this healthy wine,repeatability and the recovery were tested.The results showed that this method was stable and feasible.Wine samples of Ginseng prepared to use to relieve physical fatigue function experiments.The levels of Serum Urea Nitrogen in each dose group were lower than those in the control group,The difference between the high dose group and the blank group was statistically significant;high dose group can enhance liver glycogen reserve capacity of the mouse,increase the length of swimming.In swimming test,the area under the curve after swimming was not statistically significant compared with the control group,but there was a downward trend,showing that the test sample had alleviate physical fatigue function.Toxicological evaluation of this anti-fatigue wine was studied.The value of MTD in Acute toxicity test was higher than 21 g/kg BW,belonging to non-toxic level.The results of Micronucleus test,Ames test,sperm deformity test were negative,30 days feeding experiment found no adverse effects.The results showed that this healthy wine was safe and reliable.
