Setaria italica is one of the traditional crops in China,and it is also one of the main crops in the north of China.Its genome is small and diploid.And it has a high collinearity with rice,sorghum,maize,et al.As a result,it is one of the model plant of the grass genome,as well as a model plant for studying C4 plants.Based on setaria italica,this study was divided into two parts.The first part was to study the extraction conditions of proteins and the antioxidant effect of protein hydrolysate in setaria italica.The second part was the bioinformatics research of superoxide dismutase gene family.To srudy the extraction conditions of millet protein,extraction time,solid-liquid ratio of extractant and substrate,concentration of extractant was researched.The result showed that the best time of extracting millet proteins was 3h.The best solid-liquid ration of extracting gliadin and glutenin was 1:14 and 1:16.The best concentration was 4% NaCl to extract globulins,70% ethanol to extract gliadins,0.5% NaOH to extract glutenins.To carry out the enzymolysis part,computer simulated hydrolysis was used first.Then four type of hydrolysis was executed: trypsin hydrolysis,pepsin hydrolysis,combined hydrolysis of thermolysin and trypsin,thermolysin and pepsin.The result of computer simulated hydrolysis showed that 72 peptides of 65 types were gained using trypsin,while 264 peptides of 122 types were obtained using pepsin.Combined of thermolysin and trypsin,7436 peptides of 2239 types were gained.And 6344 peptides of 2198 types were gained with the combination of thermolysin and pepsin hydrolysis.he result of computer simulated hydrolysis also indicated that the effect ofpepsin was better than trypsin,while the combination of thermolysin of trypsin was better than the combination of thermolysin and pepsin.The experiment of radicals scavenging proved the computer simulated hydrolysis,and also showed the capacity of scavenging DPPH of combined hydrolysis was much better than using a single enzyme.Based on the genome database of millet,superoxide dismutase(SOD)was analyzed in bioinformatics.The result showed that there were eight genes in SOD family of millet,contributed on 6 chromosomes.The length of amino acids was in the rage of 56 to 334 aa.The number of exons was among 1 to 9.Three domains and 5 motifs were analyzed.Compared with other species,phylogenetic tree revealed that SOD proteins in setaria italica were similar with oryza sativa and sorghum bicolor.The results of the study provided the theoretical basis to study the function of SOD genes and proteins.
