Millet(Setaria italica) is one of China's traditional food crops. Its planting area is broad, and it has high nutritional value and antioxidant capacity. Sucrose synthase, the key enzyme of sucrose metabolism, can affect the synthesis of major amounts of antioxidants and antioxidant capacity in plants. Due to, this topic will be divided into two parts to study.The first part is the antioxidant effect of foxtail millet extract research, the second part is the bioinformatics research of millet sucrose synthase gene families.In this test, five varieties millet were used to study the antioxidant content of total phenol, flavonoids, polysaccharide and alpha tocopherol and the antioxidant capacity by using spectrophotometric method and HPLC method. The result showed that the total phenol, flavonoids,polysaccharide and alpha tocopherol content ranged from 79.31 to 36.99mg·100g-1, 16.21 to 38.51 mg·100g-1, 3.35~15.93mg·g-1 and 0.712 to1.531 mg · 100g-1 respectively in millet extract. The IC50 of DPPH free radical scavenging, OH- free radical scavening and total antioxidant capacity ranged from 0.09 to 0.24mg·mL-1, 0.41 to 0.74 mg·mL-1 and 0.98 to 2.35 mg·mL-1 in alcohol extract. Compared with the alcohol extract, the water extract of DPPH, OH-free radical clearance rate and total antioxidant capacity was weak. The total antioxidant capacity in Zhang zagu3 was slightly higher than the other four varieties. Correlation analysis results showed that the DPPH radical scavening was correspond with polyphenols,alpha tocopherol and flavonoids. OH- free radical scavening was the correlation of polyphenols, flavonoids. The correlation of total antioxidantcapacity and alpha tocopherol is bigger. Antioxidant capacity of alcohol extraction is stronger than water extract. In five millets, Zhang zagu3 had the stronger antioxidant activity.Sucrose synthase(SuS), a kind of protein in plants to converse sucrose,plays a very important role in the metabolism of plants. Based on millet genome database platform, we used bioinformatics to study millet SuS gene families. Results showed that SiSuS(Setaria italica Sucrose synthase)gene family had nine members which were uneven distributed on 5chromosomes. The length of amino acid sequence ranged from 809 to 1088 AA. The number of exon ranged from 10 to 16. The isoelectric point showed that most of the SuS family's proteins had weak acidity. According to the analysis of amino acid sequence of foxtail millet, Si SuS had nine conservative motifs. Compared with other species,evolutionary tree analysis revealed that millet, Oryza sativa and Sorghum bicolor Moench of SuS protein were similar. There were 27 genes in evolutionary tree which can be divided into 4 groups. There were 9 of orthologous genes and 1 of the homologous genes in that tree. The results of the study provided the theoretical basis and evolutionary origin to understood the function of millet SiSuS genes.
