Glucosinolate(GS)are significant metabolites related with insect and disease resistance of plants.They widely exist in the model plant Arabidopsis thaliana and cruciferous plants.According to different precursor amino acids used for their biosynthesis,glucosinolates can be divided into three types: indolic glucosinolates,aliphatic glucosinolates and aromatic glucosinolates.Indolic glucosinolates are involved in series of plant regulatory activities,including the morphogenesis,the defense of pathogen,herbivores and insects,as well as the biosynthesis of IAA and Camalexin.The synthesis of indolic glucosinolates in Arabidopsis thaliana can be regulated by three important transcription factors,namely MYB34,MYB51 and MYB122,which provide significant practical and theoretical views for research in this process.Plant hormones,such as abscisic acid(ABA),jasmonic acid(JA),salicylic acid(SA),gibberellin(GA)and ethylene(ET),also have effects on the biosynthesis of indolic glucosinolates.The analysis of promoter sequences of MYB34,MYB51 and MYB122 through Plantcare indicates that the three transcription factors may be regulated by plant hormones.In order to investigate their expression patterns and interactions with different hormones,three promoters have been linked with GUS reporters.The constructions are as follow: PMYB34::GUS,PMYB51::GUS and PMYB122::GUS.All these constructions were inserted into Arabidopsis by using the inflorescence infection method with agrobacteria.Transgenic lines were used in analysis of expression pattern and changes after hormone treatment.The main conclusions of this paper are as follows:(1)Acquire PMYB34::GUS,PMYB51::GUS and PMYB122::GUS plant expression vectors and corresponding transgenic Arabidopsis linesOn the basic of p CAMBIA3300 NLS-GUS,we inserted promoters of the three genes.Acquired three plant expression vectors PMYB34::GUS,PMYB51::GUS and PMYB122::GUS,into Arabidopsis through agrobacterium,and finally got the transgenic Arabidopsis.(2)Obtain the expression patterns of MYB34,MYB51 and MYB122 transgenic Arabidopsis linesIn terms of MYB34,it was expressed in the roots,hypocotyl.After maturation,the expression was in the roots,petioles,flowers and siliques.In terms of MYB51,it was mainly expressed in the roots of seedlings,the expression level in new leaves is higher than that of cotyledons.After maturation,MYB51 is mainly concentrated in the leaves,but also found in lateral roots,flowers and siliques.In terms of MYB122,it expressed in vascular tissue and roots of seedlings.After maturation,it expressed in roots,leaves,flowers and siliques.(3)Expression patterns of three genes after hormone treatmentExpression of MYB34 was induced by ABA,SA and Me JA,whereas ACC and GA induction were not significant.Expression of MYB51 was induced by SA and ACC,while ABA and Me JA could inhibit MYB51.Expression of MYB122 was induced by SA,GA and Me JA,whereas ABA could inhibit MYB122.The expression patterns of MYB34,MYB51 and MYB122 involved in the regulation of indolic glucosinolate synthesis under hormones were elucidated by GUS staining,providing reference dates for the future study in indolic glucosinolate transcription factors and plant resistances.
