Triterpenoid is one of the most important biological active components of the Hedyotis chrysotricha, with strong and wide pharmacological effects of antioxidant anti-aging, antibacterial and anti tumor, and broad prospect of development. This experiment is to investigate the extraction, separation and antioxidant, antibacterial activities of the Triterpenoid in Hedyotis chrysotricha and so on for further research and development of the triterpenoid in hedyotis chrysotricha. The main research results of this study are as follows:(1) Triterpenoid was isolated from Hedyotis chrysotricha by using ethanol. The best parameters of technology were compositely using single-factor test, orthogonal design and Response Surface Methodology. The study was involved in extraction frequency, extraction temperature, extraction time, solid-liquid ratio and concentration of ethanol. Experimental results showed that the best extraction condition of triterpenoid was: extraction once, 1:25 for solid-liquid ratio, extraction temperature 70 ?, 1.8h of extraction time, 89% ethanol to whole extraction system.(2) Triterpenoid was isolated from Hedyotis chrysotricha by using Ultrasonicassisted extraction technique. The best parameters of technology were compositely using single-factor test, orthogonal design and Response Surface Methodology. The study was involved in ultrasonic temperature, ultrasonic time, ultrasonic frequency,ethanol concentration and ultrasonic power, establishing a multiple quadratic equation of the yield of triterpene and influence factors, Experimental results showed that the best extraction condition of triterpenoid were: extraction once, 1:20 for solid-liquid ratio, ultrasonic extraction temperature 72 ?, 39 min of extraction time, 62 w of Ultrasonic power,.(3) By means of TLC identifying Triterpenoid, using chromatography to separate two Triterpene extractums: ursolic acid and oleanolic acid, then identify the structure by nuclear magnetic resonance spectroscopy.(4)The experimental results of the antioxidation of Triterpenoid: The reducing capacity: VC > Ursolic acid > Oleanolic acid > Triterpene extractum; Scavenging free radical activity of DPPH size : VC > Ursolic acid > Oleanolic acid > Triterpene extractum; Hydroxyl free radical scavenging activity: VC > Ursolic acid > Oleanolic acid > Triterpene extractum.
