This thesis aims to design a fast, accurate and low cost tuberculosis diagnostic tool which can help preventing tuberculosis threatening the world health. Serodiagnosis is a relatively fast and inexpensive tuberculosis diagnostic method. Two usual serodiagnostic methods, enzyme-linked immunosorbent assay and microbead suspension assay are both based on labeled detection. We think it is possible to further reduce the test complexity and cost by applying a label-free detection system in serodiagnostic assays. On the other hand, porous silicon(PSi) has attracted more and more attention because of its advantages. Much work has been reported on PSi biosensing field, which makes it possible to apply PSi biosensor in real world bio-detection. In this article, we designed a label-free antigen-antibody diagnostic tool based on PSi for serodiagnosis. This tool was used to detect the interaction between 16 kDa antigen and anti-16 kDa antibody, which are meaningful in tuberculosis diagnosis. The experimental results indicate that our biosensor has high sensitivity and specificity. The highest sensitivity of our sample can reach 0.05nm/μg/mL and the lower detection limit is 1 μg/mL.
