Digital microscope, which consists of auto-focus technology, theaccurate control of the object stage position and the image mosaicstechnology, is of fundamental importance to the medicinal and technology,molecular and cell biology, environmental biology and other fields.Firstly, the paper suggests a unified approach, which combines DFD andDFF to get the clear image. The paper applies the modified DFDalgorithm to roughly focus and DFF to accurately focus. Aimed at theclear image of whole slice, the paper suggests a focal distance estimationmethod for every view field:the Inverse Distance to a Power inlow-power and Region Harmony in high-power,which are applied toestimate the focal distance of the whole view field. Secondly, in order toimprove the dynamic performance of the auto-focusing system,Fuzzy-PID control is introduced. The paper simulates the stage drivenbased on the Fuzzy-Control by the MATLAB/Simulink to provide areference. And then, The Fuzzy-PID control is implement based on Single-chip system in hardware design. Finally, Aimed at misaiming themicroscope image efficiently, the paper improve the SIFT to be suitablefor the microscope based on the known displacement.At last, the software and hardware, by which the system realize theresearch content above are introduced in detail. Comprehensiveexperiment are design to validate the feasible of auto-focus technology,the accurate control of the object stage position and the image mosaicstechnology for the digital microscope.
