Objectives:To analysis the correlation between the single nucleotide polymorphism of rs708272 loci in cholesteryl ester transfer protein(CETP) gene with exercise induced the changes of serum lipid in obese adolescents, and investigate the molecule mechanism.Objects and methods:(1) Objects:fifty obese adolescents (23 are male and 27 are female) from north China, the average age is 14.9±3.35, both of them agreed to take part in this research. (2) Method:subjects take a month-long campaign which is based on aerobic exercise and complementary with strength and stretch training to lose weight, with unified diet; before and after the campaign the fasting venous blood was drawn for determining plasma lipid indicators and extracting genomic DNA, then use PCR-RFLP method to analysis the correlation between them; meanwhile, construct recombinant plasmid containing wild type fragment or mutant type fragment which achieved by PCR, detecting the luciferase report gene expression activity of recombinant plasmid after transfection into 293T cells.Results:(l)The distribution characteristics of the CETP gene rs708272 loci of 50 obese adolescents is:the frequency of B1B1 genotype was 0.26, B1B2 was 0.56, B2B2 was 0.18, conform to the H-W balance, can be as a representative of the group. (2) After one month aerobic exercise, B1B2 genotype obese adolescents group’s plasma TC (0.64 ± 0.51 mmol/L), LDL-C (0.51 ± 0.41 mmol/L) decreased greater than B1B1 genotype group (TC:0.20 ± 0.53 mmol/L; LDL-C:0.24 ± 0.35 mmol/L); the level of plasma HDL-C of B2B2 genptype group (0.10±0.15 mmol/L) and B1B1 genotype group (0.18±0.26 mmol/L) increased higher than B1B2 genotype group (-0.03±0.16mmol/L). (3) The relative activity of the luciferase reporter containing B1 allele (11.40±0.26) of rs708272 polymorphism was significantly lower than the B2 allele (31.19± 1.50), and both were significantly higher than the control group (3.78±0.11).Conclusion:(1) Obese adolescents who have B2 allele of CETP gene rs708272 is more sensitive to the movemwnt plan of this research. (2) The first intron of CETP gene has a positive role in the regulation of gene expression, and the mutation from B1 to B2 of rs708272 loci is a functional mutation, it can significantly increase the activity of CETP gene expression.
