| Objective:To explore the effect of adrenergic receptors(ARs)gene polymorphism on HIIT intervention effect,and to screen molecular markers of cardiopulmonary fitness and lipid metabolism training effect after HIIT intervention,so as to provide reference for formulating personalized precise fitness program.Methods:In this study,245 healthy college students(114 male and 131 female)of Han nationality who are not specialized in sports were recruited and intervened with HIIT scheme three times a week for 12 weeks.The indexes related to cardiopulmonary fitness and lipid metabolism were tested before and after intervention.DNA was extracted from venous blood and genotyped.SNPs associated with phenotypic indexes were screened by Plink 1.90 software.The differences of phenotypes among different genotypes were analyzed by independent sample T test or one-way ANOVA.According to the contribution of different genotypes to phenotype,the Genotype Predisposition Score(GPS)was calculated,and the explanatory degree of GPS to phenotype was calculated by stepwise regression analysis,and the prediction model was constructed.ROC curve was used to evaluate the accuracy of the prediction model and to find the best critical value.Results:1.ADRA2A rs1800545 locus was significantly correlateed with VO2max baseline.GPS-rs1800545 could explain 0.90%of that individual difference in VO2max baseline.2.ADRA2A gene rs553668,rs1800544,rs1800038 loci were very significantly correlated with that change rate of VO2max-VE.GPS-rs553668+rs1800544+rs1800038could explain the individual difference of 1.10%VO2max-VE training effect.3.ADRA2A gene rs1800545 and rs1800038 loci were very saignificantly correlated with that VT-VO2change rate.GPS-rs1800545+rs553668+rs1800038couldexplain 2.40%of the individual differences of VT-VO2train effect.the ADRA2A rs1800545 locus was significantly associated with VT-VO2baseline,and GPS-rs1800545 could explain 0.80%of that individual variation in VT-VO2baseline.4.ADRA2A gene rs1800545 locus were very significantly correlated with that change rate of VT-VE,ADRA2A gene rs1800038 loci were significantly correlate with that change rate of VT-VE.GPS-rs1800545+rs1800038 could explain 3.30%of the individual difference of VT-VE training effect.The ADRA2A gene rs1800545 was significantly associated with that basal VT-VE,and GPS-rs1800545 could explain 0.80%of the individual variation of the basal VT-VE.5.ADRA2A gene rs1800545 locus was significantly associated with the resting HRrest change after HIIT exercise intervention.ADRA2A gene rs1800545 and rs553668 were significantly associated with that resting HRrest phenotype,and GPS-rs1800545+rs553668 could explain 8.70%of the individual difference in HRrest.6.ADRB3 gene rs4994 locus was significantly correlated with that change rate of SBP.GPS-rs4994 could explain 5.30%of the individual difference of SBP training effect.ADRA2A gene rs553668 and rs1800544 were significantly associated with SBP baseline.GPS-rs553668+rs1800544 could explain 1.80%of the individual difference of SBP baseline.7.ADRA2A gene rs553668 was significantly associated with the rate of change of body fat,and GPS-rs553668 could explain the individual difference of training effect of 1.40%body fat.ADRA2A gene rs1800038 locus was significantly correlated with that basal body fat.The rs4994 locus of ADRB3 gene was significantly associated with baseline BMI.8.There was a significant correlation between rs1800545 of ADRA2A gene and TC change rate.The rs1800038 locus of ADRA2A gene was significantly correlated with the baseline TG.The rs1801253 locus of ADRB1 gene was significantly correlated with baseline LDL-c.Conclusion:1.HIIT can improve VO2max,VO2max-VE,HRrest and BPrest of Han college students,and then improve cardiopulmonary fitness.rs1800545,rs553668,rs1800544,rs1800038 of ADRA2A gene and rs4994 of ADRB3 gene can be used as SNP markers for the effect of cardiopulmonary fitness training;The rs1800545 and rs553668 loci of ADRA2A gene can be used as SNP markers of cardiopulmonary fitness baseline.2.HIIT can improve the fat content,BMI,triglyceride and lipoprotein of Han college students,and improve the lipid metabolism;rs553668 and rs1800545 of ADRA2A gene can be used as SNP markers of lipid metabolism training effect;The rs4994 site of ADRB3 gene and rs1801253 site of ADRB1 gene can be used as SNP markers for basal lipid metabolism. |
