In this dissertation, isolation of the effective component and harmful substances in Radix Pseudostellariae (Chinese name is Taizishen) of Jiangsu geoherbs; the fingerprinting of Radix Pseudostellariae based on activity constituent are studied. Towards these objectives,the following eight topics discussed in the thesis.1. Polysaccharide determined by phenol - sulfuric acid colorimetric method , the reference substance is glucose, the glucose content is regarded as a polysaccharide content in the calculation ,so the measured values have certain differences with the actual values, the present study seeks to different contents of polysaccharide in a quantitative comparison, evaluating the quality of the Taizishen; pseudostellarinoside is tested by vanillin - acetic acid assay, in the absence of standard, so the substance ginsenoside Rb1 is regarded as the reference, and the result will have some differences with the actual contents,this experiment seeks to different contents of pseudostellarinoside in a quantitative comparison,evaluating the quality.2. Polysaccharide is tested by HPLC/ELSD method. The glucose is standard,the purified polysaccharide is hydrolyzed to the glucose,getting the polysaccharide conversion factor. Through the conversation factor,the content of glucose is converted to the content of polysaccharide in different samples , This determination is a good method for the evaluation of polysaccharide,because of high sensitivity and good reproducibility, the experiment provides a scientific basis for the quality evaluating of Taizishen.3. The content of 22 inorganic elements (17 kinds of trace elements and heavy metals five 5) are analyzed with advanced means of ICP and microwave digestion - AFS, to accurate information the accurate information of inorganic elements of Radix Pseudostellariae is obtained. The results provide a scientific basis for the quality evaluating of Taizishen, but also inorganic elements distribution map is developed, it can identify the authenticity of Radix Pseudostellariae.4. The pesticide residue of different samples from different areas is analysed by gas chromatography, the test results show that the pesticide residues in diffent samples has certain differences, the amount of pesticide residues of Taizishen are in line with standard limits.5. A simple and reliable high performance liquid chromatographic(HPLC)method has been developed and validated for the fingerprinting of extracts from the Radix Pseudostillariae. The fingerpints of different samples are different,because of differences in plant species or variations in environmental conditions of the growth locations of the plant.For the Taizishen from same location,the fingerprints of the samples shoe closely similar fingerprints. So the HPLC fingerprinting technique has high potential in herb autheticaton application.6. Radix Pseudostellariae from different habitats were analyzed by HPLC-MS. Some characteristic peaks were identified preliminarily based on the MS spectra and literature data. HPLC-MS fingerprint of 10 main peaks was established preliminarily. It was discoveried that a small number of samples differed from others obviously. The method is reliable,accurate and can be used for quality control of Radix Pseudostellariae.7. The ecological environment of Taizishen is analyzed, preliminary results show that the climate of the main district is warm and humid, the soil is Sandy or loam ,loose fertile .The heavy metals in addition to Lead (Pb) are in line with standards and consistent with non - pollution standards. So the ecological environment of the main district is good,and the environment can meet the specific environmental requirements of the Taizishen.8. The contents on the inorganic elements of Radix Pseudostellariae were determined by ICP and AFS. The data were analysed with the SPSS11.5 software. Results indicate that the accumulation of Caã€Znã€P is high in geoherbs.There is no relationship between the concentration of inorganic elements in Radix Pseudostellariae and those in their corresponding soil. |
