In this dissertation,isolation of the effective component and harmful substances in Folium Apocyni Veneti,the fingerprinting of Folium Apocyni Veneti based on activity constituent are studied and growth of different effective material accumulation and the dynamic changes were studied.Towards these objectives,the following discussed in the thesis.1.The identification of medicinal products.Identified using traditional methods and modern identification techniques,from the medicinal properties,microscopic characteristics,thin-layer chromatography and HPLC fingerprint,etc.,set up Folium Apocyni Veneti and Its Adulterant identification methods;to the active ingredients of quantitative analysis combined with the overall analysis of fingerprint patterns,evaluation of the quality of merchandise Folium Apocyni Veneti.2.Alcohol-extract.According to Folium Apocyni Veneti chemical constituents and pharmacological study of living situation,according to "Chinese Pharmacopoeia",2005 edition of the Appendix provides a method of extract used in cold alcohol-Determination extract,as a means of evaluation of the quality of medicines.3.Using UV-VIS leaf venetum Determination of total flavonoids content,Hyperin select for reference substance to determine the content of hyperin Folium Apocyni Veneti as leaf flavonoid content in the calculation of the total target,determination of value and there is a certain difference between the true value,the experiment aimed at different places on the leaf of Apocynum The content of total flavonoids quantified for comparison,the evaluation of the quality of medicines.4.Determination by HPLC of total flavonoids quercetin and four monomer content. Select quercetin as reference substance,in accordance with the "Chinese Pharmacopoeia" (2005 version) Folium Apocyni Veneti under the total quercetin content for samples prepared with 80%methanol 50mL,reflux extraction of one hour,when the hydrochloric acid hydrolysis of extract volume of 1/10.According to this study prepared for the trial to optimize the results of goods that reflux extraction 90min×3,extract volume:volume of 9:1 hydrochloric acid to determine the best total quercetin content,so as to "Chinese Pharmacopoeia" in the total Folium Apocyni Veneti quercetin content of the amendment to provide a scientific basis.Venetum select specific leaf four known flavonoid monomers (quercetin,hyperin,quercetin glycosides vary,rutin) as reference substance,the use of HPLC method for simultaneous determination of four flavonoid monomer content.This method,accurate and reliable,high sensitivity,good reproducibility,quality standards for the evaluation of Folium Apocyni Veneti study provides a scientific basis.5.The content of 23 inorganic elements(18kinds of trace elements and heavy metals five 5) are analyzed with advanced means of ICP and microwave digestion-AFS,use the SPSS11.5 to accurate information the accurate information of inorganic elements of Folium Apocyni Veneti is obtained.The results provide a scientific basis for the quality evaluating of Folium Apocyni Veneti,but also inorganic elements distribution map is developed,it can identify the authenticity of Folium Apocyni Veneti.6.The pesticide residue of different samples from different areas is analysed by gas chromatography,the test results show that the pesticide residues in diffent samples has certain differences,the amount of pesticide residues of Folium Apocyni Veneti are in line with standard limits.7.A simple and reliable high performance liquid chromatographic(HPLC)method has been developed and validated for the fingerprinting of extracts from the Folium Apocyni Veneti.The fingerpints of different samples are different,because of differences in plant species or variations in environmental conditions of the growth locations of the plant.For the Folium Apocyni Veneti from same location,the fingerprints of the samples shoe closely similar fingerprints.So the HPLC fingerprinting technique has high potential in herb autheticaton application.8.A simple and reliable high performance Capillary Electrophoresis method has been developed and validated for the fingerprinting of extracts from the Folium Apocyni Veneti. Folium Apocyni Veneti on the implementation of the overall evaluation of the quality of medicin.9.GC-MS in conjunction with technology from different areas of the leaf venetum analysis,determination of the total ion current fingerprint to determine the total peak,and part of chromatographic peaks for the initial vesting,the initial set up a peak for a total of 14 fingerprint GC-MS information of the fingerprint,found a small number of leaf production venetum HPLC-MS fingerprint of a significant difference.This method is accurate and reliable,reproducible and can be used as the internal quality evaluation of leaf venetum basis.10.Harvesting period of different dynamic analysis of changes in modern technology, combined with quantitative analysis of active ingredient overall evaluation of fingerprint pattern of growth for different time Folium Apocyni Veneti,volatile components,and other inorganic elements in the dynamic accumulation of effective substances and Analysis of changes in laws,in order to determine the best ingredients to provide a scientific basis for harvest time.11.Climatic factors on the synthesis of medicinal substances and the accumulation of the effective impact of research.Study climatic factors(temperature,light hours, precipitation,relative humidity,etc.) to medicinal substances in the effective flavonoids (total flavonoids and quercetin,rutin,quercetin glycosides vary,hyperin,etc.),volatile component of the cumulative target of evaluation,using UV and HPLC,HPCE,such as the role of climatic factors on the different ingredients of change and its effective material to study the law,using stepwise regression analysis of climatic factors on the effective impact of the accumulation of material to explore the climate factor and effective accumulation of the relevance of the material.The main feature and innovation in this thesis are presented:1 To explore the different leaf phenology Folium Apocyni Veneti effective substances and changes in the dynamic accumulation,Climatic factors and the effective accumulation of the relevance of the material,for the production of high-quality ingredients to provide theoretical guidance.2 To establish the quality and safety of Folium Apocyni Veneti scientific evaluation system by Quantitative analysis of the active ingredient in combination fingerprint pattern of the overall evaluation.3 To establish the method of rutin,quercetin glycosides vary,hyperin and quercetin 4 Determination of the active ingredient at the same time by the HPLC-DAD,HPCE-DAD analysis first time.Determinate the total quercetin to optimize the experimental conditions,for the "Pharmacopoeia" in venetum leaves determination of the total quercetin content of the amendment to provide a scientific basis.4 To esstablish the HPCE-DAD、HSGC-MS finger print analytical method of Folium Apocyni Veneti in differet habit,collection period and kind.
