| Objective:Radix pseudostellariae are the tuberous root of Pseudostellaria heterpphylla (Miq.) Pax ex Pax et Hoffm of caryophyllaceae perennial herbaceous plants. Pinellia ternate are the tuberous root of Pinellia ternata (Thunb.) Breit of araceae herbaceous plants. Above are record by "China Phamacopeia"published in 2010 and generally used in different prescription in clinical.In this study, we are take the means of pharmacoanalysis, evaluate the quality of the sample collection in GuiZhou province, study on the inherent law of medicinal materials quality change between different regions sample. To develop the basic research for improve and perfect the existing medicinal materials quality control standard.Method:Through consulting a large number of literatures, the main effective components are polysaccharide, saponins and polypeptide etc in Radix pseudostellariae. Besides total acid record by "China Phamacopeia"published in 2010, guanosine and alkaloids are also effective components in pinellia. The contents of Polysaccharides and total saponins were measured by the sulphuric acid-phenol colorimetric method and the vanillin-ice acetic acid colorimetric method. Using the method of HPLC to determine the content of cyclopeptide B, to establish the fingerprint in radix pseudostellariae by HPCE. The contents of guanosine and total acid in pinellia were determined by HPLC and potentiometric titration. According to the UV-vis spectrophotometry, Total alkaloids was determined by acid dye colorimetry.Results:Using the method of HPLC to determine the content of cyclopeptide B in radix pseudostellariae, the calibration curve of cyclopeptide B was linear among the content range of 0.0404 to 1.414μg(Y=2990.4X+42.095,r=0.9997), the average content of cyclopeptide B was 0.019%, the average recovery rate of cyclopeptide B was 99.54%,RSD was 2.11%. Using the method of HPLC to determine the content of guanosine in pinellia, the calibration curve of guanosine was linear among the content range of 0.0129 to 0.129μg(Y=2415.1X+0.4655,r=1), the average content of guanosine was 0.016%, the average recovery of guanosine was 97.70%,RSD was 2.52%. When using the ultraviolet-visible light detector, the calibration curve of total saponins was linear among the concentration range of 8.96 to 40.32μg·mL-1(Y= 0.0203X+0.0272,r= 0.9998), the average content of total saponins was 0.441%, the average recovery of total saponins was 98.99%, RSD was 2.27%; Polysaccharide present a good linear relationship among the concentration range of 5.73 to 14.31μg·mL-1(Y=0.0572X+0.0641,r= 0.9994), the average content of polysaccharide was 23.968%, the average recovery of polysaccharide was 100.102%,RSD was 2.84%; the calibration curve of total alkaloids was linear among the concentration range of 2.45 to 9.80μg·mL-1(Y=0.0662X+0.1091,r=0.9995), the average content of total alkaloids was 0.007%, the average recovery of total alkaloids was 99.56%,RSD was 2.58%. Total acid in pinellia present a good linear relationship among the content range of 2 to 40mg(Y= 0.0848X-0.0026,r=1.000), the average content of total acid was 0.388%, the average recovery of total acid was 98.64%, RSD was 1.13%. The similarity of radix pseudostellariae fingerprint is all above 0.90, the precision, stability and repeatability are conform to the technical requirements of establish fingerprint.Conclusion:Through research the medicinal components in Radix pseudostellariae and pinellia mainly product in GuiZhou province, provide scientific basis for perfect the existing quality standard in the future. We find that same chemical component in different sample collect from different regions by analyze above research data, but there is obvious regional differences in content.Using the method of HPCE to establish fingerprint about Radix pseudostellariae, There is a good similarity in Radix pseudostellariae come from 15 batches different areas, similarity is all above 0.90, the main chromatographic peak is approximately consensus. Meanwhile, we also obtain typical model. |
