Objective:To explore the expression of power against PI3 K / AKT signaling pathway downstream of the critical molecular mdm2 after hypoxic-ischemic brain damage,provided experimental evidence for the clinical treatment of hypoxic-ischemic encephalopathy.Methods: The hypoxic-ischemic models were made with new-born SD rats aged 7days by colligating bilateral common carotid artery. All rats were randomly divided into control, model, sham acupuncture, electroacupuncture, electroacupuncture with antagonist and antagonist group,which subdivided 1d, 3d, 7d, 21 d time-phases. The pathology of neuron were observed by HE staining,Ultra-structure of neurons was observed using TEM, and the expression of mdm2 was tested with Western Blot.Results:Following hypoxic-ischemic brain injury, In model group cortex neurons arranged in irregular. With ischemia prolonging, a large number of neuronal degenerated, necrosis and apoptosis significantly lower, with mdm2 protein expression increasing compared with control group.In early EA group rats cortex neurons injury.But with the treatment of early electric needle, mdm2 expression levels were significantly lower compared with the same time point in the model group,apoptosis decreased, and cell injury significantly recoveried.In antagonist group rat cortex damage was the heaviest, apoptotic cells increased significantly, and the expressions of mdm2 protein were at a high level at the same time.Conclusion: Electro-acupuncture can reduce nerve injury and promote its repair and its mechanism may be associated with decreased expression of mdm2 to protect nerve.
