Merr. in the mulberry family, it has hot alexipharmic effect and dieresis, used for lung hot cough, fever polydipsia, bone steaming hot flashes, hot shower acerbity pain, damp heat, diarrhea, fever drugs ulcers and skin itchinghas and has high medicinal value. Humulus scandens resource is rich, the north and the south throughout the provinces of our country except Xinjiang and Qinghai. Now contains in local medicinal materials in the standard of Sichuan, Jiangsu, Shanghai and Fujian province and so on. Hebei province local standard has not yet contains Humulus scandens. Humulus scandens has a wide distribution of origin, quality differences between different regions is larger, the existing research shows that in Beijing and Hebei produce Humulus scandens celery apigenin-7-O-β-D-glucosidase and luteolin content is higher, but not someone made a systematic study on the quality of medicinal materials of Humulus scandens in Hebei. To this end, this paper conducted the related research of Hebei Humulus scandens medicinal material production quality.The first part Microscopic and TLC identification and determination of moisture, total ash and acid insoluble ash of Humulus scandens in HebeiObjective:To determinate the characteristics of microscopic identification, method of TLC identification, and moisture, total ash and acid insoluble ash content of Humulus scandens in Hebei.Method:Seting up the method of TLC identification through the sample processing method, a system, TLC plate and alkaline, temperature and other experimental conditions of humulus scandens in Hebei; Microscopic identification and moisture, total ash and acid insoluble ash content determination according to the 2010 edition of “ China pharmacopoeia â€(a) under appendix method.Results:The crystal cell, stalactite, non-glandular hairs, threaded pipe, bordered pit catheter, calcium oxalate crystals and porosity and so on were identified with microscopy from the powder of Humulus scandens in Hebei. Thin layer chromatography, thin layer plate : ilica gel G alkaline precast slab(0.4%); An agent : toluene, ethyl, formate-formic acid(5:4:1),under normal temperature.All samples had same color of fluorescence with luteolin reference substance corresponding position.The moisture ranged from 8.4% to 10.0%, the average was 9.4%, the total ash ranges from 11.8% to15.7%, the average was 13.4%, acid insoluble ash ranged from 0.4% to 2.3%, the average was 1.1% of Humulus scandens in Hebei.Conclusion:Fiber, crystal cell, non glandular hairs, threaded pipe, bordered pit catheter, calcium oxalate crystal clusters, leaf epidermal cells and porosity can be used to produce medicinal powder microscopic characteristics identification of Humulus scandens in Hebei. The method of luteolin TLC identification is established, and separation effect is good, the Rf values is moderate and the chromatographic spots is cilear, and has good adaptability and reproducibility, and can be used in the identification of Humulus scandens. Moisture, total ash and acid insoluble ash content is lower than that of “ Chinese pharmacopoeia †2010 edition(a) similar medicinal value. The second part Determination of luteoloside, apigenin-7-O-β-D-glucosidase and luteolin in Humulus scandens in HebeiObjective:To establish determination method of luteoloside, apigenin-7-O-β-D-glucosidase and luteolin the three kinds of Humulus scandens in Hebei. To comparise the content of luteoloside, apigenin-7-O-β-D-glucosidase and luteolin of Humulus scandens from different habitats, different months, female plants and male plants in Hebei.Methods:Determinating three kinds content of luteoloside, apigenin-7-O-β-D-glucosidase and luteolin simultaneously of Humulus scandens by high performance liquid chromatography(HPLC). Detection method is established by preparating of test sample solution, detection wavelength, mobile phase, optimizing chromatographic column, the precision, stability and repeatability of the method, the recovery ratio of sample investigation. Testing the different samples with the content determination method has been established.Results:System suitability conditions, Column : Agilent 5 TC-C18(250 mm × 4.6 mm, 5 μm); mobile phase : acetonitrile-0.2% phosphoric acid; flow rate : 1.0 m L / min; detection wavelength : 340 nm; column temperature : 25 ℃; Injection volume : 10 μL. Preparation of the test method:ake the No. 3 screen medicinal powder 2 g, 70% methanol 30 m L, heating reflux extraction 1 h, extracted one. According to the geographical distribution, from south to north, luteoloside, apigenin-7-O-β-D-glucosidase and luteolin content generally showed ascendant trend of Humulus scandens in Hebei. Three kinds of component content ranged in 0.0053%~0.2332%, 0.0008%~0.0521% and 0.0008%~0.0521%. According to different harvest time detection, contents of luteoloside and apigenin-7-O-β-D-glucosidase was high in Humulus scandens in July and August in Xingtai, Langfang, Shijiazhuang, Tangshan and Chengde; The content of luteoloside of Humulus scandens was high in July in Xingtai and Tangshan. The content of luteoloside was high of Humulus scandens in September in Shijiazhuang, Langfang and Chengde.According to male and female plant, contents of luteoloside, apigenin-7-O-β-D-glucosidase and luteolin of Humulus scandens in females plant were higher than male plant in Handan, Xingtai, Langfang, Tangshan and Chengde. The average content of luteoloside in females was about three times as many male strains, average contents of apigenin-7-O-β-D-glucosidase and luteolin in females was about twice as many male strains.Conclusion:The contents of luteoloside, apigenin-7-O-β-D-glucosidase and luteolin of Humulus scandens can be determinated by HPLC,the method is simple, accurate and reproducible. hree kinds of ingredients of Humulus scandens in different regions, different months, the female and male plants in Hebei is significantly different. The third part Study on HPLC characteristic spectrum of Humulus scandens in HebeiObjective:To establish characteristic spectrum of Humulus scandens production in Hebei.Methods:HPLC method.Determinating the total of absorption peak of Humulus scandens from different origin herbs in Hebei based on the chromatograms of determination and using the traditional Chinese medicine(TCM) chromatographic fingerprint similarity evaluation system(2004 edition), fitting Hebei produce medicinal materials of Humulus scandens feature maps,and evaluating the different origin samples chromatogram similarity with feature maps.Results:The method of HPLC characteristic spectrum has be established of Humulus scandens in Hebei. Compared with the fitting of feature maps.number of S1 and S3~S11 of Humulus scandens similarity were above 0.900, including Handan, Hengshui, Shijiazhuang, Cangzhou, Langfang, Tangshan, Qinhuangdao, Zhangjiakou and Chengde; Only the No.S2 of Humulus scandens similarity was less than 0.900,in other words Xingtai was low similarity of Humulus scandens.Conclusion:This experiment fitting feature maps can be used as feature maps of Humulus scandens in Hebei. |
