Humulus scandens also is known as Lara seedlings, Lara vine and so on, which is a perennial herbaceous vine and belong to Sankoh Humulus genus. It is a dried whole plant medicine, which can eliminate the heat and toxins, etc.It is used for tuberculosis hot flashes, gastroenteritis, dysentery, colds, fever, dysuria, pyelonephritis, acute nephritis, cystitis, urinary stones from oral administration;Its external use include carbuncle furuncle swollen poison, eczema, snake bites, etc.Humulus polyphenols also is called as Humulus tannin, which is Widespread in the the Humulus roots, leaves and fruit, whose content is only after cellulose, hemicellulose and lignin, is secondary metabolites and natural organic compounds. The main chemical composition of the whole plant are luteolin, glucoside, choline, asparagine, volatile oils, tannins, resins and other substances, leaves contain the cosmos glycosides and vitexin, fruit contains humulone and lupulone. Polyphenols includ phenolic acids and flavonoids, so it can be seen most of pharmacological compounds are polyphenols in Humulus scandens.Polyphenols is as the effective composition of Humulus scandens, which has great research value, but there are few reports about the Humulus polyphenols and it is rarely used in the drug development stage, so the development of the Humulus scandens resources have not done enough to seriously make drug development stagnated.This article studied the accumulation law of the Humulus scandens inclusions, the extraction method of polyphenol and optimization of extraction process, measured the Humulus scandens inclusions and polyphenol content in the different growth stages and the antioxidant activity of the polyphenols alcohol extracts, providing a reference for the future comprehensive development and utilization of Humulus scandens. So draw the following main conclusions:1、By measuring the content of the crude protein, crude fat, crude fiber and crude ash in different harvest time, I found accumulation law of the Humulus scandens nutrition, which suggestted that the direction of inclusion use was to consider as an indicator considering harvest time of the Humulus scandens,, the study found the best harvest time should be determined the most appropriate harvest was the mid-August to late, three kinds of the inclusions in four reached to the most quantity,The content of crude protein, crude fat, crude fiber was18.01%,2.98%,15.64%, crude ash content was up to14.97%in early August. 2、Humulus polyphenols were extracted by conventional, ultrasonic and microwave method, and in accordance with polyphenol yield to do the single factor experiment, through processing experimental data to obtain the best extraction solvent and the best conditions of the three different conditions. The best extraction solvent is ethanol; the best single factor of the conventional method extraction conditions:concentration of ethanol was60%, solid to liquid ratio was1:15, the extraction time was30min; the best single factor of the ultrasonic method extraction conditions:concentration of ethanol was60%,solid to liquid ratio was1:15, extraction time was50min; the best single factor of the microwave method extraction conditions:ethanol concentration was30%, solid to liquid ratio was1:10, extraction time was30s, the microwave power was80W.3、A comparative study was to choose the best extraction solvent and the best extractant concentration in the different extraction methods in Humulus polyphenol extract, and the best process parameters of each method were optimized. Conventional extraction (25±3℃) at room temperature, the best optimum parameters was as follows:extracted with70%ethanol two times and30min each time, the solid-liquid ratio was1:15; the best optimum parameters of the ultrasonic extraction was as follows:extracted with60%ethanol two times in200W ultrasonic power and1h each time, the solid-liquid ratio was1:15; the best optimum parameters of the microwave extraction method was as follows:with40%ethanol2times and30s each time in80W microwave power, solid-liquid ratio was1:20.4、By measuring the polyphenol content of different harvest stage,I found the accumulation law of the polyphenols in its growth stage, polyphenols was an important component indicators to determine the best harvest time, found the middle of August is the best harvest time.5、A comparative study of the alcohol extracts through the different optimal extraction process of polyphenols was antioxidant activity, and carried out a comparative evaluation, found that the polyphenol extract had good antioxidant capacity:the scavenging effect on hydroxyl radicals and superoxide anion of alcohol extracts of the ultrasonic method and microwave method was significantly higher than the alcohol extracts of the conventional method, the scavenging effect on hydroxyl radicals and superoxide anion of the alcohol extracts of the ultrasonic method was slightly worse than the microwave method, so in Humulus polyphenol extraction, ultrasonic and microwave method is the most appropriate, but the microwave method is more time-saving and efficient.The main innovation of this reaserch was to study the accumulation law of the four inclusions in Humulus scandens harvest stage,and determined the best single factor of the different extraction method conditions and the optimal process parameters of the different methods, had researched the polyphenol accumulation low in Humulus scandens harvest stage, and the polyphenol antioxidant of these different methods and the best harvest stage was studied, enriched the study of the biological activity of Humulus scandens content for the future Humulus caould be take into the deep development phase of the foundation.
