Objective: To explore the effects of Angelica sinensis volatile oil on SHR about the mechanism in the development of essentional hypertension, the following different indicators were measured, such as blood pressure of SHRs,the morphology of myocardial tissue, the expression of vasoactive peptides of RAS in serum, and miRNA expression profiling of myocardial tissue in SHRs and providing scientific foundation for Angelica sinensis with the pharmacological mechanism.Methods: All the SHRs of 8 weeks were divided into model group, captopril group, Angelica group, Angelica captopril group, same-aged wistar rats as control group, then the systolic blood pressure of the tails were measured in different groups before and after treatment. After treatment of 4 weeks, all the rats were were put to death. The changes of pathological sections in myocardial tissues were observed. In addition,renin、AngII and Ang(1-7) were measured with ELISA. MiRNA expression profiling of myocardial tissue were measured by Affymetrix mi RNA 4.0 Array. KEGG analysis was used to identify target genes and real-time polymerase chain reaction was used to validate the effect of miRNA expression profiling. Data was processed with spss21.0.Results: 1 After treatment of 4 weeks, compared with model group, the systolic blood pressure of Angelica group decreased, and the difference was statistically significant(P<0.01).2 Studies via pathological sectioning for myocardial tissues of rats in different groups showed differences. Compared with model group, myocardial fibers of Angelica group were thinned and organized with sharply defined stripes structure. 3 Compared with model group, rennin level of Angelica group decreased, and the difference was statistically significant(P<0.01). Compared with model group, AngII level of Angelica group decreased, and the difference was statistically significant(P<0.01). Compared with model group, Ang(1-7)level of Angelica group decreased, with no statistical significance(P>0.05). 4 Compared with model group, Angelica group owned 29 different miRNAs(P<0.05), with 13 miRNAs up-regulated and 16 miRNAs down-regulated, and captopril group owned 28 different miRNAs(P<0.05), with 15 miRNAs up-regulated and 13 mi RNAs down-regulated and Angelica captopril group owned 25 different miRNAs(P<0.05), with 4 miRNAs up-regulated and 21 miRNAs down-regulated. KEGG analysis was used to identify target genes. miR-19a、 let-7i、miR-181 c were related to Insulin signaling pathways, let-7i 、miR-181a、miR-455 were related to VEGF signaling pathways and miR-122、miR-181a、miR-200 b 、 miR-181 c 、 let-7i 、 mi R-19 a were related to apoptosis signaling pathways(P<0.05).Three miRNAs(miR-155, let-7i and mi R-122) were validated with real-time polymerase chain reaction, demonstrating a relative good consistence with the result of mi RNA array.Conclusion: Angelica sinensis volatile oil can decrease blood pressure in SHRs and can regulate mi RNA expression profiling of myocardial tissue and can protect myocardial tissue, with the possible mechanism of Insulin signaling pathways and VEGF signaling pathways, also with the possible mechanism of reducing rennin and AngII in serum.
