Objective: We observed the protective effect of Angelica volatile oil on vascular endothelial injury induced by hypertensive rats which were produced by L-NAME,and explored the protection mechanism.Method: Male Wistar rats 60,SPF grade,10 weeks age,weight(180±20)g.The normal group contained 8 rats which were randomly selected.The hypertensive rats were produced by gavaging L-NAME(60 mg/kg/d)5 weeks.The experiment groups were divided into model group,nifedipine group,and high,medium,low dose groups of Angelica volatile oil(n=8).Treatment groups were gavaging each drugs and the normal group,the model group were gavaging physiological saline once a day lasting 8 weeks.The experiment content included observing rats physiological condition.Measured each rats body weight and systolic pressure changes.Intraperitoneal injection of 10% chloral hydrate anesthesia,blood was collected by abdominal aorta.Detected serum NO,NOS,ET-1,vWF and TM.Pathological changes of thoracic aorta were searched by HE.The changes of thoracic aorta ultrastructure were observed by scanning electron microscopy.Circulating endothelial cells was measured by flow cytometry.Used qPCR technology to detect vWFmRNA,TMmRNA in artery.Evaluating the CRT qualitative and quantitative results used by Immunohistochemistry and Western blotting skills.Results: 1)The normal group rats physiological condition were well.The spirt and body weights of model group were bad(P<0.05),Each treatment groups spirt and body weight were improved at different time(P<0.05).2)Systolic blood pressure in model group were increased greatly comparition with normal group(P<0.05).Systolic blood pressure of each treatment groups were decreased at different level,and systolic blood pressure of the high dose groups of Angelica volatile oil in 4 weeks were decreased improtantly.After 8 weeks,systolic blood pressure were basically down to normal in high dose groups(P<0.05),but the low dose groups of Angelica volatile oil had little effect.3)The level of serum ET-1 was higher in model group than that of normal group,and the level of serum NO,NOS were both reduced in model group(P<0.05).The level of serum ET-1,NO,NOS in treatment groups were improved at different level(P<0.05).4)The level of serum TM,VWF were higher in model group than that of normal group(P<0.05),but the level of serum TM,VWF were both cut down in high,medium,low dose groups of Angelica volatile oil(P<0.05).5)The result of HE in normal group showed rat arterial intimal structure was normal,no thickening,smooth surface,endothelium continuous complete without protrusions,defects and cell adhesion.The result of HE in model group showed rat arterial intima thickening significantly,luminal narrowing,endometrial structure disorders,endothelial bulge,defect.The result of HE in each treatment group was improved at different level,but the low dose groups of Angelica volatile oil had little effect.6)The result of rat thoracic aorta scanning electron microscopy in normal group showed the thoracic aorta vascular endothelial structure is complete,smooth surface,neatly arranged,spindle-shaped,cell-free adhesion and migration.In the model group,the thoracic aorta scan showed that the thoracic aortic endothelium injury was associated with severe exfoliation,subcutaneous tissue exposure,rough surface,structural disorder and flipped,showing "insect-like" defect,cell adhesion migration.Each treatment group was improved at different level,and nifedipine group and high dose group was better than the middle dose group,moreover the low dose groups observed endothelial rupture,defects and collapse.7)The number of circulating endothelial cells in peripheral blood of rats in model group were increased obviously than those of the normal group.The result showed endothelial severely injure.The number of circulating endothelial cells in peripheral blood of rats in nifedipine group and high,middle dose group were both lower(P<0.05).8)Relative expression of TMmRNA and vWFmRNA in model group were increased compartion with the normal group(P<0.05).Relative expression of TMmRNA and vWFmRNA in experiment groups were cut back(P<0.05).9)Compared with the normal group,the expression of CRT activity in the intima of the thoracic aorta was higher in the model group(as indicated by the arrows in the model group).Compared with the model group,the CRT activity of the thoracic aorta CRT was decreased.10)Western blotting showed the level of CRT in rat abdominal aorta at model group was increased than that of normal group(P<0.05),the level of CRT in rat abdominal aorta at each experiment groups was cut back(P<0.05).Conclusion: Angelica volatile oil can reduce the blood pressure of hypertensive rats which were produced by L-NAME,which the potential mechanism were related to regulating the serum level of NO,ET,NOS,vWF,TM,vascular endothelial vWF,TM expression,and reducing the number of circulating endothelial cells,thus Angelica volatile oil plays a protective role on vascular endothelial injury in high blood pressure.The potential mechanism of protecting the endothelium may be related to the down-regulation of CRT expression by antagonizing Ca2+,relieving endoplasmic reticulum stress,and CRT in the vascular endothelium.
