| Objective: To explore the possible target and mechanism of treating gout arthritis using Gout Honeysuckle Granules by observing the effect of them on related factor, gene and protein expression in gouty arthritis rat model. At the same time to further clarify its efficacy and provide new research ideas and theoretical basis for the treatment of the disease.Methods: 60 SPF SD male rats were randomly divided into the blank control group, model control group, western medicine group, high dose group, middle dose group, and low dose group of Gout Honeysuckle Granules. To increase rat serum uric acid by adenine and amiodarone and inject uric acid sodium into ankle joint cavity on this basis to replicate the model of gout arthritis. Intervene to the model rats with Gout Honeysuckle Granules. After 7 days, detect serum uric acid levels in rats by uric acid enzyme, XO activity in liver by Spectrophotometry, expression of IL-1β, TNF-α,MMP-3 and LP-PLA2 in rat ankle joint soft tissue by ELISA, expression of HSP70 in rats by immunohistochemistry and the expression of HSP70 m RNA in rat model.Results: 1 Gout Honeysuckle Granules decreased rat serum UA levels significantly. Compared with the model group, there was a significant difference in high dose group(P<0.01), and there was difference in middle dose group(P<0.05);Gout Honeysuckle Granules decreased rat liver xanthine oxidase activity significantly.Compared with model group, there was difference in high and medium dose group(P<0.05); 2 The expression of IL-1βlevels were significantly reduced in all dose groups of Gout Honeysuckle Granules compared with model group. The difference was statistically significant(P<0.01); Same about TNF-α in high and medium dose group; 3 Gout Honeysuckle Granules decreased MMP-3 levels significantly.Compared with the model group, there was a statistically significant difference in high dose group(P<0.01), statistical difference in middle dose group(P<0.05); Gout Honeysuckle Granules decreased LP-PLA2 levels significantly. Compared with the model group, there was a statistically significant difference in middle dose group(P<0.01), statistical difference in high and middle dose group(P<0.05). 4 Gout Honeysuckle Granules significantly increased expression of HSP70 in ankle soft tissue of rats. Compared with the model group, there were statistically significant in high and medium dose group(P<0.01), statistical difference in low dose group(P<0.05); All dose groups of gout Honeysuckle Granules increased the expression of HSP70 m RNA. The difference was statistical compared with the model group(P<0.05).Conclusion: Gout Honeysuckle Granules can reduce significantly the BUA of model rats, activity of Liver XO and levels of MMP-3, TNF-α, LP-PLA2 and IL-1β in ankle soft tissue. Meanwhile, they can induce the high expression of HSP70 m RNA.According to this, Gout Honeysuckle Granules treating patients with GA are maybe though reducing BUA level in model rats and alleviate the inflammatory reaction to achieve effect of clearing heat toxin, detumescence and relieving pain. |
