Study Of Action Mechanism Of JWSM Against Gout

Through the experimental model of hyperuricemia, acute gouty arthritis, paw swelling, pain models by using modern scientific methodsm,The anti-gout activity were detected about JWSM through studing the gout-related indicators.Hyperuricemia experiment modol were made by yeast extract or uricase inhibitors in mice and determined serum uric acid levels in it, to research the effect of JWSM. The method of Adenine and ethambutol was used to cause rats'hyperuricemia model, and serum uric acid and XOD were measured to observe the effect of JWSM on hyperuricemia rat model; MSU were injected at the subcutaneous of rats right foot cause inflammation, anti-inflammatory activity were detected by measuring the volum of inflammatory toe;Acute gouty arthritis rabbit model were established according to McCarty DJ's method,,;and content of IL-1, IL-8 and TNF-αof synovial fluid, were detected to observe the on model of rabbit acute arthritis .Hyperuricemia experiments show that JWSM can significantly drop serum uric acid level,Meanwhile can significantly droped xanthine oxidase. The rabbit acute gouty arthritis experiment show that JWSM can notably reduce the content of inflammatory fators,ease inflammatory symptoms and prevent further developpment of inflammation. Rats paw swelling experiment show that JWSM can significantly inhibited rat paw swelling rate. acetic acid writhing test and tail-flicking test showed that writhing times and tail-flick latency were deleased , JWSM have effect in analgesia .JWSM have good therapeutic effect in gout's experimental modol. Its mechanism is mainly in lower blood uric acid, inhibit the xanthine oxidase activity, reduce the inflammatory factors in acute gouty arthritis, so as to achieve the effect of anti-gout. And at the same time, JWSM were showed having some effect in analgesia and anti-inflammatory that were was good for gout.
