Objective:Gout is a disease that involve uric acid and purine metabolic disorders cause elevated serum uric acid levels,then following causes urate deposition,leading to gouty arthritis or uric acid nephropathy.In order to study the therapeutic effect of traditional Chinese medicine on gout,this study used traditional Chinese medicine prescription,Tongfengxiao2 as the research object,discuss the effect of Tongfengxiao2 on purine metabolism in hyperuricemia model mice and the effect of inflammatory factors in rat models of acute gouty arthritis.Method:1 Observe and study the effect of Tongfengxiao2 on purine metabolism in hyperuricemia model mice:35 clean ICR male rats,weighing(20 ±2)g,were randomly divided into blank group,model group,positive drug group,high-dose group of Tongfengxiao2 and low-dose group of Tongfengxiao2,7rats per group.Intragastric administration for 28 days.After 1h of the last administration,replicate the model animal,the method is intraperitoneally inject mice with 0.02mL/g hypoxanthine(except blank group)to increase serum uric acid in mice to form a hyperuricemia model.One hour after the model was replicated,the mice were excised and blood was collected,remove the eyes from the mice and collect blood.After the blood was left to stand,centrifuge serum with a 3500rpm centrifuge,and measure the serum adenosine deaminase(ADA),guanine deaminase(GDA),and serum creatinine(Scr)levels of every rat in each group.2.Observe and study the effect of Tongfengxiao2 on inflammatory factors in acute gouty arthritis model rats:36 clean male SD rats,weighing(180 ± 20)g,were randomly divided into blank group,model group,positive drug group,high-dose group of Tongfengxiao2,middle-dose group of Tongfengxiao2,and low-dose group of Tongfengxiao2,14 rats per group.After intragastric administration for 18 days,except the blank group rats,perform model replication in other groups of rats,method is injection of sodium urate(MSU)in the ankle joint to induce acute gout arthritis,4 days after successful replication,obtain the rat ankle joints.The ankle joint was fixed in formalin solution for 1 week and placed in EDTA solution for 3 months to soften the ankle bone mass.Afterwards,repair,dehydrate,embed,make paraffin-embedded specimens,section for the ankle joints,perform HE staining and immunohistochemical staining.Result:Determine and analyze the concentrations of ADA,GDA,Scr in serum of mice in each group and the average optical density of IL-1?,IL-6 and TNF-?in rat joint cavity.Observe the rat joint HE staining with a microscope.Then obtain he following results:1.Tongfengxiao2 can reduce the serum ADA,GDA and Scr content in hyperuricemia model mice.2 High-dose group of Tongfengxiao2 and positive drug group can significantly reduce the expression of IL-1 and IL-6 in the joint cavity of rats with acute gouty arthritis,and at the same time inhibit the positive expression of TNF-?.3 The HE staining in microscope of Tongfengxiao2 high dose group are not have inflammatory infiltration,the thickness of synovial tissue reduced.Conclusion:1 Tongfengxiao2 can help reduce uric acid production by reducing the content of ADA and GDA in mice serum,and at the same time it has a certain degree of protective effect on the damage of renal function in HUA model mice.2 Tongfengxiao2 and positive drug group can significantly inhibit the positive expression of IL-1?,IL-6 and TNF-? in rats with acute gouty arthritis,thereby inhibiting the inflammatory reaction in rat joint sac.3.Tongfengxiao2 can inhibit ankle's inflammatory response of acute angioarthritis model rats by inhibit nflammatory infiltration and synovial tissue thicking.
