Objective:Observe the drug safety of Zangyao Ershiwuwei Ercha Ointment,and the effection to expression of Interleukin-4 and Interferon-γ which is in rat model of chronic eczema.To discuss the mechanism of Zangyao Ershiwuwei Ercha Ointment in treat chronic eczema.Method:The frist-Experiment:On safe medication of Zangyao Ershiwuwei Ercha Ointment, Give drug to rats back normal skin and artificially damaged skin by different doses which is greater than human body,s normal dose.To observe the rat skin,diet,weight,feces,eye and mucous membrane,breathing and mental status, behavior and death after give drug.At the same time,observe skin with or without erythema,edema for acute toxicity experiment,skin allergic experiment and skin irritation experiments.The second-Experiment:The research of Zangyao Ershiwuwei Ercha Ointment on experiment of IL-4,IFN-γ in serum of chornic eczema model in rats.According to random number table method rats can be divided into blank control group and model group.Using DNCB establish the rat model of chronic eczema,and then divided into five group,model group,Zangyao Ershiwuwei Ercha Ointment high dose group,Middle dose group,Low dose group and Hydrocortisone butyrate cream group.Every group make percutaneous drug treatment for 10 days.During this period,observe treatment area,skin changes,record score and the pathological histology change.Using ELISA method to detect the rat serum interleukin-4 and Interferon-γ level changes,to explore the mechanism of Zangyao Ershiwuwei Ercha Ointment treatment of chronic eczema.Results:The frist-Experiment:Through acute toxicity experiment,skin allergic experiment and skin irritation experiments,found that Zangyao Ershiwuwei Ercha Ointment has no medicine toxicity, allergies and skin stimulation.The second-Experiment:(1).Zangyao Ershiwuwei Ercha Ointment can effectively improve the pathological organization performance of rat chronic eczema,such as extend of trochanterellus, stratum spinosum hypertrophy, hyperkeratosis, lymphocyte infiltration and intercellular.After ten days treatment the lesions skin get significant improved. Middle dose of Zangyao Ershiwuwei Ercha Ointment and Hydrocortisone butyrate cream has equal curativeeffect,which is better than other groups.(2).The impact on the level of interleukin-4 and Interferon-γ in rat chronic eczema serum..Zangyao Ershiwuwei Ercha Ointment can make interleukin-4 level lower,and improve the level of Interferon-γ to adjust the immune balance of T cell. And middle dose group has equal curative effect to Hydrocortisone butyrate cream.Conclusion:Zangyao Ershiwuwei Ercha Ointment has high drug safety coefficient,after external use found non--toxic, irritation and hypersensitivity.At the same time can make effectively improve to rats chronic eczema lesions shin.After treatment interleukin-4 level lower,and Interferon-γlevel get higher.so the mechanism of treatment to chronic eczema is associated to the level of interleukin-4 and Interferon-γ,Change and the banlance of T cell.
