Objective:The comparative study:The effect of bloodletting and electroacupuncture on CUMS model rats behavior and the expression of hippocampal NOS. Method:Normal male rats 48 were given CUMS combined isolated and evaluated by the Open Field Test. After the success of the model to copy, we used the treatment of electroacupuncture (once a day,14 days) and bloodletting (3 times per week,2 weeks) for rats. When the treatment of 7,14 days were coming, the Opening Field Test was taken to evaluate the therapy effect of bloodletting and electroacupuncture. After the end of the experiment, we used the WB, PCR and immunohistochemical to detect the expression of hippocampal NOS. Result:Behavioral:during the Opening Field Test, the movement distance and the number of center areas to explore of model group significantly reduced, compared with normal group. Treatment for 7 days, the movement distance of bloodletting group rather than the electroacupuncture increased significantly (P< 0.05), compared with model group. Treatment for 14 days, the movement distance and the number of center areas to explore of the bloodletting and electroacupuncture group were significantly increased (P<0.05) compared with model group, the increased movement distance of bloodletting group was more than electroacupuncture (P<0.05). NOS:The expression of nNOS was decreased significantly in model group (P<0.01) compared with normal, bloodletting and electroacupuncture could promote the expression of nNOS (P<0.05). The expression of eNOS in model group was significantly reduced (P<0.01) compared with normal, bloodletting and electroacupuncture could improve the expression of eNOS (P<0.05), the bloodletting was better than electroacupuncture on improving the expression of eNOS(P<0.05).The expression of iNOS and NT in model group were increased significantly (P<0.01) compared with normal group, bloodletting and electroacupuncture reduced the expression of iNOS and NT(P<0.05).For the influence of NT, the bloodletting group was significantly superior to electroacupuncture(P< 0.05).Conclusion:Bloodletting (3 times per week,2 weeks)have an advantage over electroacupuncture(once a day,14 days) on reducing the depression-like behavior caused by CUMS stress and the expression of hippocampal NOS.
