Objective:To study the frequencies of allele and genotype of TM (thrombomodulin) in the populations of Guangxi and the patients with sepsis, and to explore which SNP may be associated with the incidence of sepsis, and to analyze its role in the occurrence of sepsis.Methods:The polymerase chain reaction-single base extension (PCR-SBE)technique and DNA sequencing were used to examine the polymorphism of TM in 172 patients with sepsis and 132 health volunteers in Guangxi. The distribution frequencies of the genotype and allele were compared among different groups.In order to evaluate whether the polymorphism of TM were associated with the occurrence and the developmentent to sepsis. Enzyme linked immunosorbent assay was used to measure the serum concentration of TM. Serum TM was significantly increased (P<0.05) in patients with sepsis compared with health controls.Results:In this study, one hundred and seventy two patients with sepsis and one hundred and thirty two control people were enrolled. Compare to the control group, The frequencies of CT genotype and TT genotype were higher in rs1042580C/T polymorphism of TM gene, the difference between them was statistically significant (P< 0.05). However,The frequencies of C end T allele rs1042580C/T polymorphism in the case group were higher than that in t control group [OR=0.818,95%CI(0.54-1.231) P> 0.0: Meanwhile,Compared with the control group, the frequencies of the other s SNPs in TM gene did not change significantly in the case group.Conclusion:Rs1042580C/T polymorphism in TM polymorphism may be associated with the occurrence and the developmentent to sepsis. No effect of rs1962C/T、 rs3176123G/T、rs6082986G/A、rs6113909G/A and rs1042579G/A polymorphism to the release of serum TM was observed in patients with sepsis.
