Objective:To study fasudil (Fas) intervene on adriamycin(ADM) induced myocardial injury in perioperative period in rats. The aim of the research was to provide certain experiment basis in myocardial protection for cancer patients who accepted neoadjuvant chemotherapy during perioperative anesthesia.Method:A total of 32 healthy female SD rats (weight 180±20g) were randomly divided into four groups with 8 rats each group as follows:(1) NS group (group A):Physiological saline (NS)were intraperitoneal(IP) injection with dose lOml/kg in 10:00 AM at the 1,3,5,7,9,11th day, a total of 6 times; And the same dose of NS were IP injection in 10:00 AM every day from the beginning of the 14th day in all rats of group A, and a total of 6 times. (2)Adriamycin group(group B):ADM were intraperitoneal(IP) injection with dose 2.5mg/kg (diluted to 10ml/kg before injection) in 10:00 AM at the 1,3,5,7,9,11th day, a total of 6 times; And NS were IP injection with dose 10ml/kg in 10:00 AM every day from the beginning of the 14th day in all rats of group A, and a total of 6 times. (3)ADM+Fas2.5mg/(kg-d) group (group C):A total of 6 times before adminstration were same to group B, and fas were IP injection with dose 2.5mg/(kg-d) (diluted to 10 ml/kg with NS before injection) in 10:00 AM every day from the beginning of the 14th day in all rats of group A, and a total of 6 times. (4) ADM+Fas10mg/(kg-d) group (group D):A total of 6 times before adminstration were same to group B, and fas were IP injection with dose 10 mg/(kg-d) (diluted to 10 ml/kg with NS before injection) in 10:00 AM every day from the beginning of the 14th day in all rats of group A, and a total of 6 times. All blood samples were collected as following points:T1(before the experiment), T2 (the 3th day), T3 (the 15th day), and T4(the 20th day, and anesthetized with 10% chloral hydrate for all rats). Myocardial tissues were detected in all anesthetized rats. The serum NT-pro BNP, H-FABP and cTnI levels were detected with ELASA method from separated serum, and myocardial mitochondrial swelling degree also were detected with ELASA method; The content of ATP, ADP and AMP in myocardial tissues were detected with high performance liquid chromatography (HPLC), and myocardial injury pathological grading degree were observed by light microscopy.Results:(1) The change of serum NT-pro BNP, H-FABP and cTnI levels in four groups of rats as follows:There were all not statistically significant difference in four groups of rats at T1 (P>0.05). There were all increased significantly in four groups at T2 (P<0.05), but there were not statistically significant difference that compared between group B, group C and group D at T2 (P>0.05). There were not statistically significance in group C and group D that compared with group B at T3 (P>0.05), and no significant difference that compared between group C and group D(P>0.05). There were significantly decreased in group C and group D that compared with group B at T4 (P<0.05); But serum NT-pro BNP, H-FABP expression level were statistically significant increased in Group C that compared with group D(P<0.05). (2) Comparison of content of ATP, ADP and AMP in four groups of rats as follows:The ATP content in group B were all lower than that in other three groups (P<0.05); But ADP content were not statistically significant difference that compared between the four groups (P>0.05); The AMP content in group B were all decreased significantly that compared with other three groups (P<0.05), and AMP content in group A were all increased significantly that compared with group C and group D(P< 0.05), but there was not significant difference that compared between group C and group D (P>0.05).(3)Myocardial mitochondrial swelling degree in four groups of rats as follows:Mitochondrial swelling degree, in group D increased significantly that compared with group A (P< 0.05), and that in group A, group C and group D were all decreased significantly than that in group B (P<0.05), but that in group C increased significantly than that in group D(P<0.05). (4) Myocardial pathologic grading in four groups of rats as follows: Myocardial pathology score in group B, group C and group Dwere all significantly increased that compared with group A (P<0.05), and that in group C and group D were all significantly decreased that compared with group B(P<0.05).Conclusion:(1) Myocardial injury induced by ADM in rats.(2) The serum NT-pro BNP, H-FABP and cTnI levels were decreased, the content of ATP, AMP were increased, myocardial fiber dissolving, degeneration, necrosis degree of ease, and myocardial injury degree were reduced after Fas intervention. It was suggested that Fas intervention could provide certain protective effect on ADM induced-myocardial injury in rats.
