ObjectiveThrough the contrast of liver-Yang hyperactivity in patients with hypertension and non liver-Yang hyperactivity in patients with hypertension and normal human serum micfoRNAs (miRNAs) expression profiles of different definite hypertension of liver-Yang hyperactivity and miRNA between whether there is a correlation, to explore the hypertension of liver-Yang hyperactivity in serum of patients with whether there is a specific expression of a miRNA or several miRNAs.Methods-1.Collect the Fasting serum of patients with early hypertension or non-primary hypertension but not taking antihypertensive drugs and healthy persons in our medical center and cardiology clinics,then divided them into three groups,namely liver-yang hyperactivity group and non-liver-yang hyperactivity group and normal group.2.Respectively from the elbow vein blood 5ml, stored in dry tubes without anticoagulant, 3000r/min centrifugal 10 minutes, serum was collected and frozen -30 degrees. The extraction of RNA in serum by Trizol LS method, according to the miRNeasy Mini Kit manual, using adsorption column to concentrate the RNA.3.Using Solexa sequencing method for gene sequencing of the above three sets of samples sample,detecting differentially expressed genes between liver-Yang hyperactivity group and non-liver-Yang hyperactivity group, liver-Yang hyperactivity group and normal group, non-liver-Yang hyperactivity group and normal group.4.Using online prediction software miRWalk2.0 to integrate the differentially expressed genes between liver-Yang hyperactivity group and non-liver-Yang hyperactivity group, liver-Yang hyperactivity group and normal group,to selecte the miRNAs associated with liver-Yang hyperactivity.5.Filtering out the detection of expression of miRNAs in the groups using real-time fluorescent quantitative PCR, including its concentration and total amount.ResultsFound that the expression of miRNAs of liver-Yang hyperactivity group and the normal group, there were significant differences (P<0.01); liver yang hyperactivity group and the non liver-Yang hyperactivity group compared miRNA expression also exist significant differences (P<0.01); the expression of non liver-Yang hyperactivity group compared with normal group miRNAs no difference (P>0.05). liver-Yang hyperactivity group and the non liver yang hyperactivity group miRNA concentration compared with the presence of significant differences (P<0.01); the concentration of liver-Yang hyperactivity group and the normal group of miRNAs also exist significant differences (P<0.01); non liver yang hyperactivity group and normal group miRNA concentration had no significant difference (P >0.05). liver-Yang hyperactivity group and normal group:were detected miRNA 1037, which differentially expressed miRNAs have 653 (P<0.05),223 down regulated miRNAs expression, expression of upregulated miRNAs 430; non liver yang hyperactivity group and the normal group:a total of detected miRNA 963. The differentially expressed miRNAs have 573,256 downregulation of miRNA expression, expression upregulated miRNAs 317; liver-yang hyperactivity group and the non liver yang hyperactivity group:a total of detected miRNA 957. The differentially expressed miRNAs have 720, expression down regulated miRNAs 277, expression upregulated miRNAs 433. Results miR-126 were found in highly expressed genes. Quantitative fluorescence showed that the expression of liver-Yang hyperactivity group miR-126 non liver yang hyperactivity group and the normal group (P< 0.01) were significantly higher than those in the.ConclusionHypertension of hyperactivity of liver-Yang form associated with miRNAs,and miR-126 may be one of the specific miRNAs.
