Objective: To explore the clinical basis of hospital preparations TengFuJiangYaTablets treating Liver-yang Hyperactivity Syndrome in Essential Hypertension.Toobserve its influence on the blood pressure and probe into action mechanisms, in order toprovide the reference for the research and development of new antihypertensivedrug,TengFuJiangYa Tablets.Methods: In the Clinical basis study, we collected patients with Liver-yangHyperactivity Syndrome in Essential Hypertension, to enter concerned data into the TCMinheritance system (V2.0.1)for data mining. In animal experiments, SBP,DBP,MBP weredetected by non-invasive rat tail method when SHR were at conscious state.We tookenzyme-linked immunosorbent assay to detect the state of Endothelin(ET), nitricoxide(NO), AngⅡ, Renin, Aldosterone(ALD) in SHR serum. And serum metabolites inSHR were detected by high performance liquid chromatography time of flight massspectrometry (HPLC-MS) technology.Results: In clinical basis study, the most frequently used drug were uncariarhynchophylla and semen raphani, frequency of93times and75times respectively.Uncaria rhynchophylla and semen raphani were the commonly used pharmaceuticalcompositions. By correlation degree analysis, the confidence coefficient of uncariarhynchophylla and semen raphani was0.96. Core combinations and new prescriptions alsecontianed uncaria rhynchophylla and semen raphani. In animal experiments, a single theTengFuJiangYa Tabelts did antihypertensive effect(P﹤0.05), and continuous dosing fourweeks have antihypertensive effect(P﹤0.05), especially the high dose group. Besides, ET,Renin, AngⅡ, ALD in serum of SHR(P﹤0.05) declined evidently. And the state of NO trend to rise in serum of SHR(P>0.05). The results of PCA have shown thatmetabolic pattern is different between SHR and WKY.The metabolic pattehrn changedafter the intervention of TengFuJiangYa Tabelts,and high dose group was closer to thenormal control group in space,and its effect was better than the middle dose group and lowdose group.Conclusion: The combination of Uncaria rhynchophylla and Radish Seed is themost commonly used drug pair treating Liver-yang Hyperactivity Syndrome in EssentialHypertension. There is clinical basis in treating the disease with TengFuJiangYa Tablets.TengFuJiangYa Tablets have effectively antihypertensive function, presenting the obviouslimitation and concentration-response relationship. The medicine can balance ET/NO,tosome extent, improve the endothelial function, effectively inhabit the activity ofrenin-angiotensin-aldosterone system(RAAS), and improve the metabolic pattern of eachgroup. The study provide the reference for the research and development of newantihypertensive drug,TengFuJiangYa Tablets.
