Background:Spleen deficiency syndrome refers to the status of digestive system dysfunction, clinically visible eating less with abdominal distension, loose stool, lassitude, less vigor and lazy words, yellow or pale complexion, pale tonguewith whitish coating, slow and weakpulse etc. As can be seen from recent literatures, more research tends to be associated with the pathogenesis of spleen deficiency syndrome and gastrointestinal hormone secretion, demonstrating a series of gastrointestinal symptomsin a variety of factors. Our research reviewsthe understanding and development of modern medicine and Chinese medicine on spleen deficiency syndrome as well as general situation of massage therapy for digestive system diseases in recent years, then we study on regulating the function of gastrointestinal hormones in New Zealand rabbits with spleen deficiency by means of Pushing Manipulation with One Finger (PMOF).objective:This paper intends to adopt cold diarrhea method to make spleen deficiency New Zealand rabbit animal model. To observe the effects of different frequencies of Pushing Manipulation with One Finger(PMOF) on serum biochemical and gastrointestinal hormone secretion of spleen deficiency New Zealand rabbits in order to further study on the relationship between amount of manual stimulation and the reinforcing and reducing methods as well as to provide a certain experimental basis for elucidating the mechanism and clinical application of PMOF.Methods:The general level of 50 New Zealand rabbits, weighing from 1.5 to 2.5Kg, male and female, provided by Experimental Animal Center of Guangzhou University of Chinese Medicine. Adaptive feeding for 7 days, randomly selected 8 as Blank Control Group (Group K) and the other 40 make spleen deficiency models. After modeling, the models were divided into 5 groups. One group was randomly selected as Model Control Group (Group P) and the remaining 4 groups were divided into A, B, C, D group, for the intervention of PMOF. The remaining 2 for practice spare exercises. Using senna, rhubarb, mirabilite preparation of the Bitter Cold Purgation animal model of spleen deficiency. After successful modeling, the A, B, C, D group were implemented with 4 frequency bands as50-100,101-150,151-200,201-250 (Beats/min) respectively. Group K and P does not intervene. According to the methods of taking points in Experimental Acupuncture, we take Zhongwan and Tianshu. Treatment lasts for a total of 10 days. After that, the model animals were observed in general. Biochemical indexes of serum were measured by automatic biochemical analyzer and the content of gastrin in blood was measured by radio immunoassay.Results:1. After modeling, all groups showed decreased body weight, appetite reducing anddirty hair color etc. (P< 0.05). After the manual therapy, the treatment groups had different degrees of recovery. Among them, Group B with a frequency in 101-150 (Beats/min) is the most obvious in terms of weight. The improvement about Group C and Group D, compared to the Model Group were significantly different (P< 0.05). There was no significant difference between the Group D at the frequency of 201-250 (Beats/min) and the Model Control Group (P> 0.05).2. After intervention, behavioral score in each group varying degrees of recovery. Group B faster recovery,there was statistically significant difference compared with group P (P< 0.05). Group A and C as the frequency away from group B, the degree of improvement gradually decreased, compared with group P, the difference was not statistically significant (P> 0.05). D group no significant improvement, and further tends to decrease, compared with the control group, the difference was statistically significant (P< 0.01).3. The changes of serum biochemical indexes in each model group after the models were made(P< 0.05). After manual therapy, the serum biochemical indexes of all groups tended to recover. Among them, the K ions showed the difference between groups. The recovery rate of A group to B group was gradually increased. Among them, Group B with a frequency in 101-150 (Beats/min) is the most obvious (P< 0.05) while Group B to Group C tended todecline. The improvement level of GroupC to Group D was further reducing. There was no significant difference between Group D and the Model Control Group (P> 0.05). After manual therapy, group differences on sodium and chloride were not significantly obvious (P> 0.05).4. Compared with the Control Group, the Model Group GAS had significant changes (P< 0.01). After manual therapy, the GAS in each group tended to recover, expressing of serum K ions is approximately consistent. However, Group Dwith high frequencies continued to rise after the treatment with no significant difference (P> 0.05).Conclusion:1. This experiment confirmed that the three-stage the Bitter Cold Purgation of spleen deficiency rabbit model could successfully reflect theclinical symptoms of spleen deficiency syndrome, so that the modeling method is more perfect.2. The effects of Na and Cl ions on the serum of rabbits were not significantly affected bythe method of PMOF. The effect of intervention on the internal environment of New Zealand rabbits was not significant. The PMOF has certain security, Meanwhile, it showed that animal itself has the ability of environmental homeostasis, and it has a certain degree of self-healing tendency.3. comprehensive analysis the behavior of each intervention group, the improvement of serum GAS and the level of serum K ion, we think that the best optimum frequency band of this experiment should be between 101 Beats/min to 150 Beats/min(Group B). Group A and C equally have thevalue of intervention. Meanwhile, we think that 201 Beats/min to 250 Beats/min(Group D)have no significant effect on spleen deficiency syndrome. Prompt exist the best intervention frequency values on POMF with spleen deficiency syndrome, will need to be further follow-up test.
