Objective:Endodontics and oral periapical disease is a common disease. The most common and effective treatment is root canal therapy. The principle of RCT is to prevent periapical disease by removing most infections in root canal mechanically, killing germs inside chemically, filling the root canal, and implementing crown closure. The key of root canal treatment dependents on the appliance of the root canal sealers. iRoot SP is a new bio-ceramic materials, which posses biocompatibility, antimicrobial activity, X-ray radiopacity and alkalinity. This study purpose was to provide a theoretical basis for how iRoot SP could be applied clinically by application investigating its effect on apical sealing performance in company with different filling technique.Methods:A sample was selected of 80 human teeth with single root canal extracted during the past two months. Molars inclusion criteria:no caries, apical development completed. Those teeth were randomly distributed into 4 groups(A、B、 C、D), each of of equal size of 20. These teeth root canal were prepared with ProTaper in the method of crowm down to F2 size, and then choose different materials and techniques to get the root canal oburated. Group A, obturation of root canal were done with gutta-percha+AH-Plus sealer through warm vertical compaction; Group B, root canal were obturated with iRoot SP sealer through warm vertical compaction; Group C, root canal were obturated through a single tip method with iRoot SP root canal sealer; Group D, received no obturation. After root canal obturation, all specimens under constant temperature box for a week, when root canal obturation material completely hard solid, immersing in 2% methylene blue dye solution, and were decalcified with nitric acid, then underwent dehyration of gradient ethanol, at last under salicylic acid methylester in the solution to make transparent specimen processing production. Stereoscopic microscope measuring root penetration length, using SPSS 13.0 to do the variance analysis on data used, at a 5% level of significance.Results:1.The apical micro leakage length which obturated with gutta-percha+AH plus sealer by using warm vertical compaction is(0.81±0.27)mm, significantly different from the control group (p<0.5);2.The apical micro leakage length which obturated with iRoot SP by using warm vertical compaction is (0.75±0.23) mm, significantly different from the control group (p<0.5);3.The apical micro leakage length which obturated with single cone obturation with iRoot SP is (0.97±0.12)mm, significantly different from the control group (p<0.5);4.The difference between AH plus sealer and iRoot SP by using warm vertical compaction was no statistically significant(p<0.5);5.The difference between iRoot SP by using warm vertical compaction and obturation with single cone obturation with iRoot SP was no statistically significant(p<0.5);Conclusion:1. Warm vertical compaction obturation techniques have been widely used with its high sealing capability in root canal treatment.2.The selection of root canal sealer is one of the key factors in 3D tightly filled root canal, iRoot SP is similar to AH-Plus as root canal sealer in apical sealing performance, thus iRoot SP can be used as a new root canal sealer used clinically.3.Single cone obturation could be used for root canal treatment when there are difficulties for operation in some situations, such as patients with limited opening degree or molars with deep locations.
