Pu’er tea is China’s top ten famous tea in the history, the main producing areas are Xishuangbanna, Baoshan, Lincang, Pu’er District of Yunnan province. Pu’er tea can not only make people happy, but also make people healthy. Tea polyphenols (TP) is a general designation of polyphenols in tea. There are a lot of ingredients that are unique to the tea and these substances together constitute the unique flavor and efficacy of tea. Pu’er tea can decrease blood lipid level, lower blood pressure, anti atherosclerosis, anti-cancer, bactericidal anti-inflammatory, anti-aging, and so on. Tea polyphenols is one of important function components. Gout is a disease caused by purine metabolic disorder, which is characterized by hyperuricemia (HUA) and seriously affects the health and quality of patients’lives.In this study, we used Oxonic acid potassium salt manufacturing hyperuricemia/gout mouse model, and studied the effect of Pu’er tea polyphenols on lowing serum uric acid level and its antioxidation in HUA mice.1. The response surface analysis of tea polyphenol extraction processBy regression equation analysis, we obtained the best process conditions of the highest yield of tea polyphenols, as follows:extraction time of 0.97 h, extraction temperature of 89.42℃, solid-liquid ratio of 3.93%, ethanol concentration of 74.52%.2. Establishment of the HUA mice modelTo establish the HUA model, a total of fifty mice were continuous given injection with oxonic acid potassium salt (300mg/kg) for 7 days. It inhibited the uricase activity of mice and elevated serum uric acid levels in mice.3. Studies on the effects of PTP to lower uric acid in HUA mice.All mice were divided into the five groups,10 mice for each group. OA intraperitoneal injection was given at 9:00 am per day, followed by intraperitoneal injection of tea polyphenols or allopurinol one hour later, continuous injection for 8 days. The control group was injected with the same volume of 0.9% normal saline at 9:00 am and 10:00 am. respectively.At the end of the eighth day, these mice were killed for 1 hours after the injection by taking eyeball blood, and they were dissected at once. Analyses of uric acid content and XOD enzyme activities in serum and liver in HUA mice showed that Pu’er Tea polyphenols are able to decrease uric acid levels in HUA mice.4. Studies of PTP on the antioxidant effect in HUA mice.The serum SOD activity, GSH-Px activity and SOD activity, GSH-Px activity and MDA content of liver tissue homogenate were measured for each group. Results showed that PTP had anti oxidative effects, which could eliminate free radicals in vivo.5. Liver and kidney biopsy of HUA miceThe HUA mice were dissected to take the kidney tissue and liver tissues to make paraffin section. We found the damage of renal tubular structure in HUA mice. Slight morphological variation was also observed in liver cells of HUA mice.
