Objective:To observe the protective effection and research mechanism of Xinmailong injecton (XML) on myocardial ischemia reperusion(I/R) injury in Rats model.Methods:The level of SPF 60 successful acute myocardial ischemia rat models were randomly divided into control group,I/R group and XML group(Different drug concentrations 50mg/ml,100mg/ml,200mg/ml,400mg/ml);The control group was continuously given KH nutrient solution in 2 hours and the other group was given respective different drug concentrations or KH nutrient solution by Langendorff isolated heart perfusion method after ischemia 30mins.The following indicators were detected:Hemodynamics include HR, LVDP and ±dp/dtmax; the myocardial enzymes include CK, CK-MB, Tn-I and MYO, myocardial histopathology the myocardial infarct size were observed; the apoptosis protein expression of Bcl-2 and Bax were detected by immunohistochemistry, immunofluorescence and western blot.Results:Compared with I/R group.XML group could protect myocardial systolic and diastolic function from ischemia injury; can decrease the activities of CK.CK-MB Tn-I and MYO; can reduce myocardial cell injury and shrink area of myocardial infarction; Besides, can decrease the protein expression of Bax, increase the expression of Bcl-2.Conclusion:XML can enhance heart myocardial contractility of isolated heart of the model rats with acute myocardial ischemia,improve the function of ischemic heart,and has a protective effect on myocardial reperfusion injury after acute myocardial ischemia.
