Object: To investigate the effect of Aiweixin on the heart function, arrhythmia, LDH, formazan content and myocardial apoptosis of isolated rat heart during ischemia and reperfusion. Methods: Lengendorff homoisothermy-isopiestic pressure perfused model was employed to determine intraventricular pressure, arrhythmia and coronary flow of isolated rat hearts in MedLab bio-information clollection and treatment system. LDH and Myocardial viability were measured at 550 nm by spectrophotometrically assay. Observation of myocardial ultrastructure by TEM; Determination of apoptosis by SABC immunohistochemistry. Results: Ischemia /reperfusion significantly increased left ventricular end-diastolic pressure (LVEDP), decreased maximal rise/fall rate of left ventricular pressure (±dp/dtmax) and rate-pressure product (RPP) in the isolated rat hearts. Aiweixin (5ml/L) significantly attenuated the inhibitory effects induced by ischemia on LVEDP,±dp/dtmax and RPP (P<0.01). Aiweixin (5ml/L, 10ml/L, 20ml/l) had no effect on the left ventricular end-systolic pressure (LVESP) and coronary artery flow(cf), the result had no significant difference(P>0.05). Aiweixin decreased the content of LDH in the ischemia myocardium (P<0.01). Aiweixin obviously down regulated the apoptosis index (p<0.01), and decreased the mitochondrion injury degree in ischemia cardiocyte . Conclusion: Aiweixin increased the heart function of isolated heart during ischemia/reperfusion; promoted the stability of cell membrane; inhibited the apoptosis of cadiocyte.
