| Objective:This work was to clear whether the miR-200 and miR-19 can be used as high sensitivity and strong specificity miRNA markers in peripheral blood of patients with non-small cell lung cancer;analyze the relationship between the gene expression level and the clinical pathological parameters in non-small cell lung cancer patients with peripheral blood and clinicopathological parameters;check the expression level of peripheral blood miRNA can monitor tumor recurrence and metastasis, therapy and response criteria evaluation, and the carcinoma prognosis.Methods:1. In this work,The 30 cases of healthy people,30 cases of lung cancer patients without distant metastases,30 cases of patients with metastatic lung cancer are divided into three groups.Extraction of three groups of circulating tumor cells in peripheral blood of micrornas, using real-time quantitative PCR technology to miR-200 and miR-19 two gene amplification, gene detection of three sets of peripheral blood circulating tumor cells the expression of miR-200 and miR-19, comparison between groups miR-200 and miR-19 expression level.MiR-200 and miR-19 the relative expression quantity formula is:2-△△CT.2.Analysis the relationship between the expression level of miR-200 and miR-19 in peripheral blood circulating tumor cells and lung cancer clinical pathology features and the correlation of the level of serum marker.3.We will calculate the sensitivity and specificity for detection of lung cancer by the miR-200 and miR-19 and the Serum tumor marker CEA, CA125, CA153, CA199, CA242, CA724, FER, CYFRA-12, NSE and check the sensitivity and specificity of the miRNAs.4. We use SPSS 17.0 statistical software to data processing. Measurement data use nonparametric test for statistical analysis method, hierarchical data with rank and inspection, the correlation analysis with Pearson Chi Square test.Results:1. The levels of miR-200 and miR-19 in healthy people compared to lung cancer patients without distant metastases and lung cancer are different, the differences are statistical significant (P< 0.05).2. The circulating tumor cells miR-19 expression level in health groups as follows 1.70 (0.32,3.91), without distant metastases in patients with lung cancer 11.28 (1.28,38.24), and distant metastasis in patients with lung cancer 11.46 (4.55, 4.55).Compared with the healthy group, the lung cancer group, expression level higher than the healthy group,and three groups differences was statistically significant (P< 0.05). The circulating tumor cells miR-200 expression level in health groups as follows 1.06 (0.50,0.50), without distant metastases in patients with lung cancer 2.24 (0.73,0.73).Compared with the healthy group, the lung cancer group, expression level higher than the healthy group, and distant metastasis in patients with lung cancer 5.02 (2.60,2.60),and the three groups differences statistics was significant (P<0.05). The expression level of miR-200 and miR-19 is higher in patients with lung cancer, and lung cancer metastasis group is higher than transfer group.3. Joint Monitoring specificity of miR-200c and miR-19 were higher than the monitoring of lung cancer tumor markers CEA, CA125, CA153, CA199, CA242, CA724, FER, CYFRA-12, NSE, in addition to the sensitivity of CEA were higher than other tumor markers.4.Analysis the expression level and clinical pathological parameters of miR-200 and miR-19 in the three groups,found that the expression of miR-200 and miR-19 with gender,age,pathological type, TNM staging, lymph node metastasis are not related (P> 0.05).5.The expression of miR-200 levels and tumor markers in serum CEA, CA125, CA153,CA199,CA724,NSE,FER,CYFRA-12 levels have no correlation (P>0.05), and correlated with elevated CA242 levels, was statistically significant (P<0.05).miR-19 expression levels and tumor markers in serum CEA,CA 125, CA153, CA199, CA242, CA724,NSE,FER,CYFRA-12 levels are not correlation (P> 0.05).Conclusions:1.The expression level of miR-200 and miR-19 with lung cancer was over expression in the peripheral blood circulating tumor cells,and lung cancer metastasis group expression level is higher than patients without distant metastases with lung cancer.Found that detection of circulating tumor cells in the expression levels of miR-200 and miR-19 can be used in the monitoring of tumor metastasis and recurrence.2.miR-200 expression level may be affected by CA242 serum tumor markers, combination monitoring miR-200 with CA242 indicators could improve diagnostic accuracy and reduce false positive rate in non-small cell lung cancer.Joint monitoring of non-small cell lung cancer recurrence and metastasis and prognosis is more specificity, sensitivity.3.miR-200 expression levels may be affected by serum tumor marker CA242, and joint monitoring of miR-200 and CA242 indicators could improve the diagnosis of NSCLC, reducing the false positive rate, to monitor its recurrence and metastasis, prognosis more specific, sensitivity. |
