Objective:To research the difference of serum metabolites between polycystic ovary syndrome (PCOS) patients with phlegm dampness syndrome and healthy people by the detection technology of Liquid chromatograph mass spectrometer (LC-MS), and explore the pathogenesis of phlegm dampness syndrome of PCOS from the aspect of metabonomics, searching for the related biomarkers, to provide basis for clinical diagnosis and treatment of PCOS.Method:Choose 27 PCOS patients with phlegm dampness syndrome and 27 healthy people. Collect 5ml venous blood after fasting for 12 hours from the early follicular phase (menstruation 3-4 days).Set it for 30min at room temperature,after that centrifuge it with high speed, then measure E2, LH, FSH, T, PRL, DHEAS, SHBG, FINS and FPG. Preserve the remaining serum in the refrigerator in-80 ℃. After collecting all the sample of serum, take the method of the principal component analysis (PCA) and partial least squares discriminant analysis (PLS-DA) to get the metabolic fingerprinting. Pretreat and calculate the integral of the fingerprinting.Take multivariate statistical analysis of integral quantity and construct the scores plot to analyze the differences between the two groups. Choose the biomarkers of phlegm dampness syndrome of PCOS according to the analysis of different metabolic components.Results: ①The body mass index and waist-hip ratio of PCOS patients with phlegm dampness syndrome are significantly higher than those of the healthy people (P<0.01).②Two groups on LH, T, DHEAS, SHBG, FAI, FINS, HOMA-IR have the remarkable differences. ③Comparing with the level of serum metabolites in the healthy people, the level of glycine,l-methyl histidine, diglyceride, citrate, ganglioside, ethanol, alanine, hyaluronic acid and lipid L6 are lower in PCOS patients with phlegm-dampness syndrome, with statistically significant difference (P<0.05).And its level of 2-hydroxy acid, acetacetic acid, very low density lipoprotein L5, chenodeoxycholic acid, arginine, urea, stearic acid, lipid L7, lactate, glucose, cetylic acid, n-acetyl glycoprotein and galactose are higher, with statistically significant difference (P<0.05).Conclusion:Metabonomics can distinguish the healthy people and PCOS patients with phlegm dampness syndrome, and the biological markers of phlegm dampness syndrome of PCOS are obtained:glycine,1-methyl histidine, diglyceride, citrate, ganglioside, ethanol, alanine, hyaluronic acid, lipid L6,2-hydroxy acid, acetacetic acid, very low density lipoprotein L5, chenodeoxycholic acid, arginine, urea, stearic acid, lipid L7, lactate, glucose, cetylic acid, n-acetyl glycoprotein and galactose. The changes of serum metabolites in PCOS patiente with phlegm dampness syndrome may be related to glucose metabolism, lipid metabolism and amino acid metabolism.
