Objective: Study effects and mechanism of the Puerarin on prevention of osteoporosis, to explore the Puerarin mechanism of prevention and treatment of osteoporosis by observing the expression of ER and IL-1βon bone organization.Methods:Forty female SD rats,three–months-aged,were randomly divided into five groups:sham operation group(Sham); ovariectomized group(OVX);ovariectomized and small dose puerarin group(OVX+P50);ovariectomized and large dose puerarin group(OVX+P100). There were 10 rats in each group,the group 1and group 2 as the control group,the group 3and group 4 as experimental group.When animal model established, Group 1(Sham) and Group 2(OVX) were treated with Intraperitoneal injection of 1ml saline; Group 3(OVX+P50) were treated with Intraperitoneal injection of Small doses of Puerarin(50mg/kg); Group 4(OVX+P100)were treated with Intraperitoneal injection of large doses of Puerarin(100mg/kg),for up to 8 weeks. Select the lumbar 4-5 vertebral body to experiment.Observation of the change of bone organization ultrastructure use the Hematoxylin-eosin staining.Detection changes of bone mineral density(BMD), bone mineral content(BMC) use the Dual-energy X-ray absorptiometry instrument. Observation ER and IL-1β the expression of on bone organization use the immunohistochemical method.Results : 1 Dual-energy X-ray absorptiometry instrument: Bone mineral density(BMD):OVX+P50 group spine BMD increased 2.4%;systemic BMD increased2.3%. OVX+P100 group spine BMD increased 8.5%; systemic BMD increased7.4%. Bone mineral content(BMC):OVX+P50 group spine BMC increased 11.6%;systemic BMC increased 2.2%. OVX+P100 group spine BMC increased 24.8%;systemic BMC increased 5.3%.2 Hematoxylin-eosin staining: OVX group the lumbar vertebra bone trabecular changed thin and Sparse some bone trabecular breakaged,marrow cavity expanded and appeared a large number of fat cells and large without bone trabecular bone marrow tissue. Bone resorption to reduce reconstruction increased with increase of the dose after used Puerarin, the bone trabecular volume and the number increased significantly as well as the bone trabecular breakaged and the fat cells obvious decreased,Osteoporosis has been improved.3 Immunohistochemical method: the IL-1βexpression rate of positive cells increased; the ER expression of positive cells was decreased. with increase of the dose after used Puerarin, the IL-1β expression rate of positive cells decreased; the ER expression of positive cells was increasedt.Conclusion : Puerarin could reduce of bone loss in osteoporotic model. its mechanism of action is mainly achieved by reduce bone resorption and promoting bone formation. Puerarin can effective treat postmenopausal osteoporosis caused by decreased bone strength and bone mineral content.
