On the basis of systematic review of the literature on the back pain and Puerarin in Chinese traditional medicine and Western medicine , this article objectively makes a comment on the theory about the effect of Puerarin for the osteoporotic patient.The purpose of this study is to evaluate and elucidate the effect of Puerarin for the osteoporosis patient via in vitro cell cultures , animal model and clinical study.First, in the in-vitro cell study , the effects of Puerarin on bone cell viability were determined by the method of MTT assay during the time course. The enzyme activities of osteoblasts and osteoclasts were measured by the analysis of alkaline phosphatase (ALP), and total protein content. The differentiation of osteoblasts and osteoclasts was analyzed by von Kossa stain and tartrate-resistant acid phosphatase stain, respectively.The cell survival test showed the Puerarin at concentration 5μg/mL and 50 μ.g/mL, enhancing the activity of osteoblast during proliferation stage, but no effect on ALP secretion. Obviously, the effect of herb medicine on osteoblast, keep the cell in proliferation as in early stage, but delay turning to differentiation.The potential of the Puerarin on bone cell has a positive effect on osteoblastic activity by increasing cell proliferation instead of promotingmineralization and a negative effect on osteoclastic activity by inhibiting the formation of osteoclasts.Secondly, We try to prove the Puerarin be with stimulation for bony growth activity and repairing of rabbit's large bone defect.At last, using the affiliated hospital as a base, we carefully conducted a clinical observation on the 24 random osteoporotic samplings with back pain. The result of Puerarin treatment was satisfying.Due to lack of ample observing time and case, this study deserves further investigation on the objective standard and its forward curative effect.
