Objective:To investigate the effects of Puerarin on the expression of TGF-β1and Smad2/3m RNA in Osteoblasts, and to study the mechanism of action of Puerarin on the O.P(osteoporosis), from the level of molecular biology.Methods:Separate Osteoblasts from the fetal rats of mice to cultivate in vitro, to intervene in three groups of Puerarin liquid:1ug/ml,0.1ug/ml,0.01ug/ml, and set up blank control group, to detect the activity of Osteoblasts, Adopting the method of alkaline phosphatase (AKP). Using Real-time F.Q.PCR and WesternBloting to detect variations in the expression of TGF-β1and Smad2/3mRNA, with different Puerarin intervention on Osteoblasts.Results:Different concentration of Puerarin intervention can increase the activity of Alkaline Phosphatase in the Osteoblasts, depending on the concentration of Puerarin. Real-time F.Q.PCR and WesternBloting shows that the expression of TGF-β1and Smad2/3mRNA in Osteoblasts was obviously higher than the negative control group.Conclusion:The consequence Indicates that Puerarin can promote the expressions of the Smad2/3mRNA of signaling protein in the Osteoblasts and promote the secretion and synthesis of TGF-β1.As a result, to promote the bone formation and inhibit bone absorption. It would be the curative mechanism in the prevention and treatment of the osteoporosis.
