Background and objectivesAnthracycline drugs plays an important part in breast cancer adjuvant therapy. Studies has found that anthracycline drugs target on TOP2 a protein to produce cytotoxic effect and so that induce cell apoptosis effect. But anthracycline drugs have serious side effects, so we must evaluate clinical benefit for the patient before application. This experiment aims to study how TOP2 a protein express in early nonspecial invasive breast cancer, and to analyze correlation of protein expression with the pathological grade, lymph node metastasis, the expression of ER, PR and HER-2, Ki-67 etc clinical pathological data, in order to draw the optimum anthracycline drugs application for individual screening method,so as to guide the clinical diagnosis and treatment.MethodsIn this research we Selected 35 cases of early nonspecial invasive breast cancer patients from December 2014 to 2015 October in First Affiliated Hospital of Soochow University. We use immunohistochemistry(ICH) to detect TOP2 A protein expression;analyze the correlation-ship with TOP2 a protein expression and clinical and pathological data by T test and Spearman rank correlation analysis.Results1.The expression level of TOP2 a protein was significantly positively correlated with the histological grade of the tumor.2.The tumor is greater, the TOP2 A protein expression level is higher. However the TNM stage did not show a link with the expression of TOP2 A protein.3.ER and TOP2 a protein expression is correlated, ER-negative tumor tissue detected significantly more TOP2 A protein expression.4.Her-2 and TOP2 a protein expression is correlated, HER-2-positive tumor tissue detected significantly more TOP2 A protein expression.5.Triple negative breast cancer detected highest TOP2 A protein expression quantity of the four breast cancer molecular types. In addition to luminal A type(the sample size is too small to statistical analysis), in luminal B, HER-2 over expression type, triple negative breast cancer, protein TOP2 A expression increased progressively.ConclusionsTOP2a protein expression is associated with tissue differentiation, molecular typing,and with TNM stage, there is no obvious correlation, but is related to tumor size. So we recommend patients with higher histological grade, larger tumor size, molecular typing for triple negative breast cancer to prefer to use anthracycline chemotherapy drugs.
