Objective:To probe action mechanism of Yanghe Decoction on preventing knee osteoarthritisby observing its effects on IL-land TNF-a in rabbit model with knee osteoarthritis to provide the scientific theory for its clinical practice and development and to further research the pathogenesis of osteoarthritis.Methods:24 healthy New Zealand rabbits were randomly and equally divided into the Yanghe Decoction group, the model control group and the blank control group,8 rats in each group. Except for the blank control group, the other groups were given injectin of papain to copy osteoarthritis model. After modeling, the Yanghe Decoction group was given Yanghe Decoction, the model control group and the blank control group were given saline. All above trealments were done for 5 weeks and one times a days. HE and Mankin’s score were used to observe the changes of cartilage morphology. ELISA was used to detect the content of IL-land TNF-a in blood serum and synovial fluid.Results:1. Naked eye and light microscope observation of cartilage:The blank group cartilage surface is smooth,and complete. The cartilage surface of model group was rough,defective and hyperplastic,four cartilage layer hierarchical structure is not clear and the number of cells decreased. Cell distribution is not uniform has cell clustering phenomenon, tide line disorder disappear. Yanghe Decoction group between the blank group and the model group.2.Mankin’s score of articular cartilage injury:the blank group compared the other 2 groups score was significantly increased (P<0.01); Yanghe Decoction group and model group compared significant difference (PO.01);3.ELISA test showed:The content of blood serum and synovial fluid of IL-1、TNF-a in model group compared with blank group increased significantly(P<0.01);The content of IL-land TNF-a in blood serum and synovial fluid of the Yanghe Decoction group was significantly lower than model control group, data were statistically significant (P<0.01).Conclusion:1. The OA animal model was successfully simulated by using the knee joint injection of the papaya protease2. Yanghe Decoction can reduce the IL-1, TNF-α in rabbit model of OA in serum and synovial fluid;3.Yanghe Decoction can reduce the content of IL-land TNF-a in blood serum and synovial fluid of Rabbit Model with Knee Osteoarthritis to cure Osteoarthritis.
