Part one Expression and significance of intefleukin-18, intefleukin-1β in the peripheral blood of gastric cancerObjective To determine the serum levels of intefleukin-18, intefleukin-1β and helicobacterpylori infection in gastric cancer, to explore the changes of IL-18 and IL-1β in gastric cancer and its clinical significance.Methods The levels of serum IL-18, IL-1β was detected by ELISA in 324 gastric cancer patients, 40 patients with chronic superficial gastritis, 13 patients with chronic atrophic gastritis with or without intestinal metaplasia and 20 patients with dysplasia. and the Ig G of helicobacter pylori was detected by ELISA in gastric cancer. To analyze the association between serum IL-18, IL-1β and each group,and ROC curves were used to evaluate the effectiveness of IL-18 and IL-1β on the diagnostic value.Results The levels of serum IL-18, IL-1β were significant Iy higher in gastric cancer patients than that in DYS、CAG/IM 、CSG group(P<0.05). Serum IL-18 in the DYS group and CAG/IM group were significantly higher than the CSG group(P<0.01), and in the DYS group was obviously higher than that of CAG/IM group(P<0.05). The levels of serum IL- 1 beta in DYS group is significantly higher than CAG/IM group and CSG group(P < 0.05), while there was no statistically significant difference in the CAG/IM group and CSG group(P > 0.05). The serum level of IL-18, IL-1β in gastric cancer was positively correlated with TNM stage, deepness of invasion, tumor size, lymph node metastasis,and distant metastasis; they had no significant correlation with age, sex, tumor location,degrees of differentiation in gastric cancer. Hp positive gastric cancer group was higher than the Hp negative gastric cancer patients(P<0.05).Area under ROC curve(AUC) was0.837, 0.795, respectively, while AUC of combined IL-18 and IL-1β was 0.867.Conclusions The levels of serum IL-18, IL-1β were rise in chronic superficial gastritis- chronic atrophic gastritis with or without intestinal metaplasia-dysplasia- gastriccancer dynamically,and were closely related to the degree of gastric cancer. Hp infection can increase the level of serum IL-18 and IL-1β in the gastric cancer.Part two Expression and significance of mi R-6503-5p、mi R-25-3p in the serum of gastric cancerObjective To discuss the abnormal expression of mi R-6503-5p、mi R-25-3p in the serum of gastric cancer patients and its clinical signification.Methods To detect the serum expression of mi R-6503-5p、mi R-25-3p in 322 gastric cancer patients, 40 patients with chronic superficial gastritis, 13 patients with chronic atrophic gastritis with or without intestinal metaplasia, and 20 patients with dysplasia by quantitative real-time PCR(q RT-PCR) method, and the Ig G of helicobacter pylori was detected by ELISA in gastric cancer.The association between serum mi R-6503-5p and mi R-25-3p levels with the clinicopathological features of gastric cancer were evaluated.Results Compared with the CSG, CAG/IM, and DYS group, the serum levels of mi R-6503-5p, mi R-25-3p in gastric cancer was significantly higher(p<0.05), while there was no statistically significant difference in the CSG, CAG/IM, and DYS group(P > 0.05).The levels of serum mi R-6503-5p in gastric cancer was positively correlated with lymph node metastasis, tumor size, degrees of differentiation, TNM stage and deepness of invasion(p<0.01); the expression of serum mi R-25-3p was positively correlated with lymph node metastasis, deepness of invasion and TNM stage( p<0.05), they had no significant correlation with age, sex, distant metastasis and tumor location in gastric cancer(P > 0.05). The serum mi R-6503-5p、mi R-25-3p in Hp(+) and Hp(-) gastric cancer group has no statistical significance(P > 0.05). Area under ROC curve(AUC) was 0.720,0.688, respectively, while AUC of combined mi R-6503-5p and mi R-25-3p was 0.717.Conclusions Serum mi R-6503-5p and mi R-25-3p were increased significantly in gastric cancer,and they are closely related to the clinical pathological features of gastric cancer, and they may play an important role in the occurrence and development of gastric cancer.
