Purpose:Helicobacter pylori is a strong known risk factor for gastric cancer development. However only very few infected people finally develop gastric cancer. Considering the individual differences in clinical outcome, we assume that the genetic variations may play an important role in it. Previous studies confirmed that DAF was a receptor for Helicobacter pylori. In addition, DAF can also be utilized as a cellular receptor by several other pathogenic organisms. DAF involved in tumor immune escape mechanism is closely related to tumor development. But there is no report on the association between DAF polymorphisms and Helicobacter pylori related gastric cancer susceptibility. The present work aims to explore the association and screening to provide a theoretical basis for vulnerable populations selected, early treatment, and reducing the risk of gastric cancer.Methods:rsl 0864231 and rs2782837 of DAF gene in a case-control study from a Chinese population, including 489gastric cancer patients and 489 healthy controls. Polymerase chain reaction-restriction length polymorphism (PCR-RFLP) method combined with gene sequencing method was employed to genotypes. A Chi-square test was used to calculate the difference of genotype and allele frequencies for each SNP between the case and control group. Odds ratios (ORs) with 95 percent confidence intervals (CIs) were assessed for the risk allele of each SNP by unconditional logistic regression with adjustment for gender and age. MDR was used to analyse the interaction among genes and HP.Results:The rs2782837 TT genotype was associated with an reduced risk of gastric cancer(adjusted OR,0.40,95%CI,0.19-0.98,P=0.033). In subjects without H.pylor infection, the rs2782837 TT homozygote was associated with an reduced risk of gastric cancer(adjusted OR, 0.34,95%CI,0.09-0.87, P=0.042).There was no difference between rs10864231 and Helicobacter pylori related gastric cancer susceptibility.Conclusion:Our study is the first time to reveal that DAF rs2782837 was associated with reduced risks of GC in Chinese population. Moreover, Individuals carried rs2782837 TT genotypes infection were expected to have a lower risks of Helicobacter pylori infection and gastric cancer susceptibility. There was no difference between rs 10864231 and Helicobacter pylori related gastric cancer susceptibility.
