Curcumin is the most dynamic ingredient in turmeric products, it produces the anti-tumor, antioxidant, anti-inflammatory, lowering blood pressure, falling hepatic fat, protecting the liver function and other pharmacological effects. Our work is to select the positive ways of alcohol extraction process, and so using the extraction products to perform the liver protective experimental tests on mice, Ctenopharyngodon idella and Culture alburnus, laying the groundwork for the evolution of new drugs of curcumin for the prevention and handling of aquatic animal liver disease research. The main methods of the subject and the results are as follows:The extraction, alcohol concentration, the extraction, alcohol consumption, the extraction time and the number of extraction on the result of extraction volume of Curcumin and ointment collecting rate which was evaluated by orthogonal design L9(34). The effects indicated the best extraction process: 8 times the amount of 80% alcohol, underwent 3 times extraction for 1.5 hours.The mice were administered with extract powder of turmeric in different concentrations for 7 days. Then the mice were injected with 0.12% CCl4. The levels of ALT, AST in serum and activities of SOD, CAT, MDA, and GSH-PX in liver tissue were observed and the liver lesions were examined. The results were that the activities of ALT, AST and the level of MDA in extract powder of turmeric group were significantly decreased, and the activities of SOD, CAT, GSH-PX were significantly increased(P<0.05 or<0.01), and liver pathology was improved compared with the injured group. It showed that extract powder of turmeric is under a significant protective effect against CCl4 induced acute hepatic injury in mice.The Ctenopharyngodon idella model of acute hepatic injury in vivo was induced by intraperitoneal injection of CCl4 at different dosages and different injury time, results indicated that the serum level of GOT and GPT activities were found to be significantly raised in the group injected with 15.00% CCl4 with olive oil at a mass of 0.1 mil /10 g of body weight for 24 h. Then the Ctenopharyngodon idella was gavaged the agential containing different content Curcumin for 7 days, using CCl4 to make the injured model, texting the serum index, results indicated that curcumin at the content of 150mg/kg led to substantial decreases in the activities of GPT and GOT. The findings showed the potential hepatoprotective function of Curcuma in fish. In vitro, different concentrations of curcumin on Ctenopharyngodon idella primary cell culture, the purpose of the primary cell survival rate, the results showed that with the increasing of concentration of curcumin, Ctenopharyngodon idella primary cell survival rate increased gradually, reached the maximum in 400 μM, we concluded that, curcumin could suppress the grass carp primary cell apoptosis, improve its survival rate.Through different concentrations of curcumin mix feeding for 20 days, samples were collected and detected to determine efficacy experiment. Solutions indicated that 50 mg/kg of curcumin already has the result of hepatic protecting, curcumin may have a certain hepatoprotective effect on Culture alburnus. Meanwhile, low oxygen treatment to the Culture alburnus with the blank control, the IGFBP-1 gene expression of the liver samples collected at different time was observed. Results indicated that the determination of the gene at 6 h low oxygen treatments the blank group volume had increased dramatically while the curcumin group had no substantial alterations. It may indicate the curcumin promote the hypoxia stress resistance ability.
