The Study of Therapeutic Effects and Mechanism ofCurcumin on Hepatic FibrosisObjectiveTo investigate therapeutic effects of curcumin on hepatic fibrosis and discuss itsmechanisms.MethodsRat models of hepatic fibrosis were made by carbon tetrachloride. Salviamiltiorrhiza was used as positive control. Serum levels of ALT, AST, HA, LN, PC-Ⅲ,NO, TGF-β1, TNF-α, and levels of Hyp, MDA, and the SOD in liver tissue weredetected; HE and Masson staining were conducted in hepatic tissues to observepathological variations. Grade of hepatic fibrosis were evaluated according to SSSsystem. Immunohistochemical method was executed for detecting collagenalphal(Ⅰ),(Ⅲ) and PDGF-BB in liver. Activated HSC was detected byα-SMAimmuohistochemistry stain. HSC apoptosis was detected by terminaldeoxynucleotidyl transferase (TdT) dUTP nickend labeling (TUNEL) and Desminimmuohistochemistry stain.ResultsCurcumin could decrease serum levels of ALT,AST,HA,LN,PC-Ⅲ,NO,TGF-β1,TNF-α,Hyp,MDA obviously (P<0.05), which were increased in fibroticgroup; curcumin could increase the level of SOD in hepatic tissues obviously (P<0.05), which was depressed in fibrotic group; Curcumin could obviously improveliver pathological variations of fibrotic rats, and the score of hepatic fibrosis incurcumin group reduced significantly(P<0.05). Curcumin treatment could reducedexpressions of collagen alphal(Ⅰ),(Ⅲ) and PDGF-BB(P<0.05). Aactivated HSC increased significantly in fibrotic group compared with that of normal group, andfew apoptotic HSC were detected. However, less activated HSC and more apoptoticHSC were detected in curcumin treated group compared with those of fibrotic group.These effects were dose-dependent.Conclusions1.Curcumin could improve hepatic functions and heal hepatic fibrosis.2.Curcumin have the effect of anti-peroxidation. Curcumin could enhancedegradation of collagen in liver tissue, and reduce the expression of cytokine.3.Curcumin could inhibit activation and proliferation of HSC, and induceapoptosis of HSC.4.Therapeutic effects of curcumin on hepatic fibrosis maybe related toabove-mentioned mechanisms.
