[Objective]:To investigate therapeutic effects and mechanism of curcumin on experim- ental hepatic fibrosis rats and discuss its mechanisms.[Methods]:Rat models of hepatic fibrosis were made by carbon tetrachloride. Curcumin was used as positive control. Saline injection group was used as the normal control group. Serum levels of ALT,AST,HA,LN,PC-Ⅲand CⅣ;HE staining were conducted in hepatic tissues to observe pathological variations. Grade of hepatic fibrosis were evaluated. Immunohistochemical method was executed for detecting collagen alpha1(I),(Ⅲ),TGF-β1,P38MAPK and P-P38MAPK.[Results]:Curcumin could decrease serum levels of ALT,AST,HA,LN,PC-Ⅲand CⅣobviously(P<0.05),which were increased in fibrotic group;curcumin could obviously improve liver pathological variations of fibrotic rats, and the score of hepatic fibrosis in curcumin group reduced significantly(P<0.05). Curcumin treatment could reduced expressions of collagen alpha1(I),(Ⅲ),TGF-β1,P38MAPK and P-P38MAPK.These effects were dose-dependent.[Conclusions]:1. Curcumin could improve hepatic functions and heal hepatic fibrosis.2. Curcumin have the effect of anti-peroxidation, and could reduce the expression of cytokine.3. Curcumin could inhibit the expression of TGF-β1,P38MAPK and P- P38MAPK.Therapeutic effects of curcumin on hepatic firosis maybe related to above-mentioned mechanisms.
