Objective: To investigate the dynamic changes of serum TGF-β1 and CTGF in dogs with heart failure induced by transfection of Jumonji gene. Methods: the experimental animals were randomly divided into sham operation group(group A):only pacemaker implantation, no right ventricular rapid pacing stimulation. The successful model of heart failure were randomly divided into 2 subgroups(group B and group C) with 6 dogs in each group. +AAV9-EGFP group(group B): 500 EGFP L(green fluorescent protein, titer1×1012 vg) gene of the heart failure dogs were injected with the recombinant adenovirus associated virus solution, and then the right ventricular rapid pacing was continued for 14 days. +AAV9-Jumonji-EGFP group(group C): the heart failure dogs were injected with 500 L of the gene in the heart of the heart failure dogs by the injection of Mu Jumonji gene of the virus solution(titer1×1012 vg) after continued right ventricular rapid pacing 14 days. Sustained and rapid right ventricular pacing induced canine heart failure model, measurement of left ventricular ejection fraction(left ventricular their differences from, LVEF), standard heart failure mean arterial pressure, mean artery pressure(map), measures changes in two groups of HE staining of myocardium and electron microscope observation of myocardial tissue histopathology and ultrastructure, enzyme linked immune assay for detection of canine serum TGF-β1 and CTGF, and then the immunohistochemical method, RT-PCR and Western blot detection jumonji gene and protein changes.Results: 1 successful surgery and successful completion of the experiment. Model group of 12 dogs by continuous rapid right ventricular pacing in dogs after 30 days appear different degree of loss of appetite, reduced activity, quicken respirationsymptoms of heart failure, pressure(mean artery pressure, map) < 60 mm Hg(1 mm Hg = 0.133 kPa), spotlight and ejection fraction < standard heart failure mean arterial,ultra micro structure shows: muscle dissolved, mitochondria and ER long. Intercalated disc and discontinuous fuzzy torsion. 2 after injection of AAV9-Jumonji-EGFP gene into the pericardial cavity, the left and right ventricular frozen sections showed a large number of green fluorescent protein expression, which was strongly positive. The expression of Jumonji in canine left heart was detected by immunohistochemistry. The expression of a large number of brown particles was observed. The expression of Jumonji and protein in canine left ventricular Jumonji and protein showed enhanced expression of-actin and protein in the left and right ventricles. Conclusions: 1. The sustained and rapid right ventricular pacing 30 d, preparation of heart failure model was successfully heart failure +AAV9-Jumonji-EGFP groups of dogs, map and left ventricular ejection fraction(LVEF) in serum were significantly decreased, jumonji gene transfected 14 d, map and left ventricular ejection fraction(LVEF) returned to normal levels, prompt jumonji gene can significantly improve the prognosis of heart failure. 2 the serum levels of TGF-β- 1 and CTGF were significantly increased in dogs with rapid and sustained ventricular pacing, and the results were consistent with those of clinical and basic research. And after giving AAV9-Jumonji, TGF-β1 and CTGF levels decreased significantly, the combination of MAP and LVEF values decreased,the results showed that Jumonji inhibited the TGF-β1 and CTGF levels increased,reversing myocardial fibrosis, improving heart failure.
