Purpose: According to observe the express of CTGF,FN,TNF-α and IL-6 in kidney tissues of chronic renal failure rats which induced by adenine lavag, discuss the pharmacology mechanism of the Shenshuaiyin decoction delaying the renal interstitial fibrosis.Material and method:60 SD male rats(weight 180±20g) prepared by yokozawa model of chronic renal failure,while 12 rats as blank group,the remaining as modeling group.After successful preparation of the modeling group were randomly divided into model group,Shenshuaiyin group,Niaoduqing group,which respectively were given by Shenshuaiyin decoction,Chinese patent drug Niaoduqing particle per day.model group and blank group given by Na Cl per day.Dosing period lasted for 4 weeks,during which period we recorded the rats’ body weight,active status and appearance.After dosing period,we collected abdominal aortic blood for blood to check serum creatinine,blood urea nitrogen,collected left kidney tissue for observe the appearance of kidney tissue and biopsy.Check the express of CTGF and FN by immunohistochemical,TNF-α,IL-6 by ELISA,Then we statistically analyzed the result of the various results.Results:1.The express of CTGF and FN respectively increased in model group,compering with blank group have significant difference(P<0.01);Compering with model group,the express of CTGF and FN were decreased in Shenshuaiyin group(P<0.05),CTGF and FN expression level in Niaoduqing group have no significant difference(P>0.05); Compering with Niaoduqing group,the express of CTGF and FN were decreased in Shenshuaiyin group,having significant difference(P<0.05).2.The express of TNF-α respectively increased in model group,compering with blank group have significant difference(P<0.05);Compering with model group,the express of TNF-αwere decreased in Shenshuaiyin group and Niaoduqing group(P<0.01);Compering with Niaoduqing group,the express of TNF-α were decreased in Shenshuaiyin group,having significant difference(P<0.01).The express of IL-6 respectively increased in model group,compering with blank group have significant difference(P<0.01);Compering with model group,the express of IL-6were decreased in Shenshuaiyin group(P<0.01), IL-6expression level in Niaoduqing group have no significant difference(P>0.05);Compering with Niaoduqing group,the express of IL-6were decreased in Shenshuaiyin group,having significant difference(P<0.01).3.serum creatinine and blood urea nitrogen leves in model group was obviously rising than blank group(P<0.01); Compering with model group,the level of serum creatinine and blood urea nitrogen were decreased in Shenshuaiyin group and Niaoduqing group(P<0.01);Compering with Niaoduqing group,the level ofserum creatinine decreased in Shenshuaiyin group,having significant difference(P<0.05),but blood urea nitrogen level have no significant difference(P>0.05).4.Compering with blank group,model group have the most obvious pathological change,followed by Niaoduqing group,Shenshuaiyin group found little pathological change.Conclusion:Shenshuaiyin decoction may attenuate interstitial fibrosis happening and developing by reducing the expression of CTGF induced by TGF-β1and decreased the level of FN, TNF-α, IL-6.providing the basis of treating chronic renal failure.
