Objective: Picking out the same target gene of Wnt and BMP signaling pathways, and clone the target gene into the plasmid vecter of adenovirus. Finally, transfect the plasmid vecter of adenovirus into the package cell HEK293 and generate the recombinant adenovirus expressing the target gene.Methods: We pick out the same target gene of Wnt and BMP signaling pathways using cDNA microarray. We clone the picked out gene in vitro and intergrate it into the shuttle vecter of pAdTrack-CMV by restrict clonning , and recombinant it with adenovirus skeleton vecter Adeasy-1 by homologous recombination. Finally , We transfect the product into package cell HEK293 by Lipofectamine and generate the recombinant adenovirus expressing the target gene.Results: (1) Picked out the same targe gene CTGF of Wnt and BMP signaling pathways using cDNA microarray, (2) Cloned the CTGF gene in vitro and verified the true result by gene screening, (3) Intergrated the CTGF into the plasmid vecter of adenovirus and result the recombinant ade-plasmid Adeasy-CTGF, (4) Transfected the Adeasy-CTGF into the package cell HEK293 and generated the high titer adenovirus expressing the CTGF.Conclusion: CTGF is one of the most important downstream target gene both Wnt and BMP signaling pathways. We cloned the CTGF gene into the recombianant adenovirus, made a real good basic for further research.
