Electro-Acupuncture At Different Combinations Of Points: Their Impacts On The Jejunum Entero-Chromaffin Cells And 5-HT Of Functional Diarrhea Rats

Objective: By using certain parameters of electro-acupuncture to stimulate functional diarrhea rats with different combinations of points, the paper aims to observe the differences in the diarrhea indexes, the morphology and number of jejunum entero-chromaffin cells, the expression levels of 5-HT and TPH,explore the regulating mechanisms of electro-acupuncture treatment of functional diarrhea, and screen better combinations of points in treating functional diarrhea with electro-acupuncture.Methods: 20 SD rats were randomly divided into 2 groups: 10 in the control group and 10 in the model group. The control group didn’t have any treatment,while the model group received 3ml/Senna Decoction orally, 1 times/day, for14 days. At the end of the stomach filling, the two groups of rats continued to be given normal feeding, water drinking, and continuous observation for14 days. At the end of the 28 d modeling, experimental data and indicators were observed and evaluated comprehensively.50 SD rats were randomly divided into 5 groups: control group, model group,electro-acupuncture(EA) group 1, EA group 2, and EA group 3. For model group,EA group 1, group 2, and group 3, the model was made by using the above modeling method of diarrhea model. At the end of the modeling, the effects of electro-acupuncture intervention were observed. The control group and the model group were not treated with electro- acupuncture. EA groups(group 1,group 2, and group 3) were given electro-acupuncture intervention for 1 course(1 times/day, 10 days/treatment). After the course of treatment, the jejunum tissues of the rats were intercepted, and the number of jejunum entero-chromaffin cells, the secretion levels of 5-HT and the expression levels of TPH were detected.Results: Through the observation and comprehensive evaluation of weight growth rate, diarrhea index, loose stools rate, loose stools level and silver staining, the model of functional diarrhea rats made by Senna Decoction gavage proved to be successful. At the end of modeling, it could be found that compared with the control group, the number of jejunum entero-chromaffin cells, the secretion levels of 5-HT and the expression levels of TPH increased in the model group.After electro-acupuncture intervention, compared with the control group and the model group, all the three EA groups reduced the number of jejunum entero-chromaffin cells, the secretion levels of 5-HT cells, and the expression levels of TPH. Of the three groups, group 1(Tianshu with Dachangshu)had remarkable influence while the effects of group 2(Quchi with Shangjuxu)and group 3(Tianshu with Dachangshu + Quchi with Shangjuxu) were not significant.Conclusions:1.The model of functional diarrhea rats made by Senna Decoction gavage proved to be successful.2.Jejunum and ileum were the sites of senna diarrhea model and jejunum was the main site of action.3.In the process of modeling and acupuncture intervention, the number of jejunum entero-chromaffin cells, the secretion levels of 5-HT and the TPH expression levels had correlation with functional diarrhea.4.By electro-acupuncture intervention of the related acupuncture points,the number of jejunum entero-chromaffin cells, the secretion levels of 5-HT and the TPH expression levels were reduced and the signal transduction was decreased, thus the therapeutic effects were achieved. Of the three groups,group 1, namely Tianshu with Dachangshu, proved to be an effective combination of acupoints for treatment of functional diarrhea while the effects of group2 and group 3 were not significant.
