Objective:The study was to investigate the effect of acupuncture at Back-Shu and Front-Mu Points on the treatment of functional diarrhea and its effect on intestinal flora.The purpose of this study was to explore the mechanism of acupuncture treatment of functional diarrhea with acupuncture at Back-Shu and Front-Mu Points,and provide scientific basis for clinical selection and application of acupuncture treatment of this disease.Methods:Thirty Specific Pathogen Free grade Sprague-Dawley male rats were randomly divided into blank group,model group,Back-Shu point group,Front-Mu point group and Shu-Mu points group,with 6 rats in each group.The rats in the model group and the acupuncture group were intragastrically administered with a senna leaf agent to produce the functional diarrhea rat model,and the rats in the blank group were orally administered with an equal amount of 0.9% sodium chloride solution.One hour after gavage,the same day electroacupuncture Back-Shu point group(ST 25),Front-Mu point group(BL 25),Shu-Mu points group(ST 25 + BL 25),each time 20 minutes;the blank group and the model group uses the same bundling method,no needling,20 minutes;once a day,a total of 10 times.After acupuncture,rat stools were collected under aseptic conditions,and 16 S rDNA sequencing technology was used to observe the defecation,intestinal flora abundance and diversity of the rats after treatment.Results:1.Compared with the blank group,the number of defecations in the model group was significantly higher than that in the blank group(P<0.01).Compared with the model group,the number of defecations in the Back-Shu point group,the Front-Mu point group and the Shu-Mu points group was significantly reduced,and the difference was statistically significant(P<0.01).Compared with the blank group,the 24-hour fecal water content of the model group was significantly higher than that of the blank group(P < 0.01).Compared with the model group,the 24-hour fecal water content in the acupuncture Shu-Mu points group and the Back-Shu point group was significantly reduced,with statistical difference(P < 0.05),suggesting that acupuncture the Shu-Mu points and the Front-Mu point can both improve the 24-hour fecal water content in functional diarrhea rats.The 24-hour fecal water content of rats in the Back-Shu point group was higher,suggesting that acupuncture the Back-Shu point had no significant effect on the fecal water content of functional diarrhea rats.2.Compared with the blank group,the OTU of the model group was significantly reduced,and there were significant differences in the PC1 dimension and the PC2 dimension.Alpha diversity decreased significantly and Beta diversity decreased significantly.The number of Actinobacteres,Deferribacteres and Firmicutes(Ruminococcus,Faecalibacterium,Erysipelotrichaceae)increased significantly,with statistically significant differences(P < 0.01).The number of Tenericutes,Bacteroidetes,Elusimicrobia was significantly reduced,and there was a statistically significant difference(P<0.01).3.Compared with the model group,the OTU of the acupuncture Shu-Mu points group was significantly increased,and there were significant differences in the PC1 dimension and the PC2 dimension.Alpha diversity increased significantly and Beta diversity increased significantly.The number of Actinobacteres,Deferribacteres and Firmicutes(Ruminococcus,Faecalibacterium,Erysipelotrichaceae)was significantly reduced,with statistical differences;the number of Tenericutes,Bacteroidetes,Elusimicrobia increased significantly,with statistical differences.Conclusions:1.Acupuncture at the Back-Shu and Front-Mu Point can regulate the intestinal motility of rats with functional diarrhea,and reduce the amount of defecation and water content of the feces in rats.2.Acupuncture at the Shu-Mu points can improve the intestinal flora of rats with functional diarrhea: Acupuncture at the Shu-Mu points group can reduce the amount of Actinobacteres,Deferribacteres,Firmicutes(Ruminococcus,Faecalibacterium,Erysipelotrichaceae),increased the amount of Bacteroidetes,Elusimicrobia,and Tenericutes;acupuncture at the Back-Shu points group and the Front-Mu points group have no obvious regulating effect on intestinal bacteria.3.According to the existing research,it is concluded that acupuncture at the ShuMu points may reduce the intestinal motility of rats with functional diarrhea by improving the amount of Bacteroidetes and Firmicutes in the intestinal flora,so as to achieve the purpose of treating functional diarrhea.
