In 4 kinds of cultivation patterns (bed planting under forest, suspension under forest, intergrouth under forest and bed planting in greenhouse) of Dendrobium officinale as experiment materials, Comparison were made to konw the main component content in different cultivation patterns, and the protective effect of Dendrobium officinale extract on antioxidant, antibacterial and acute alcohol liver. The main results were as follows:1. The polysaccharides, amino acids, flavonoids, polyphenols and ethanol extracts content of Dendrobium officinale differs in different cultivation patterns.The polysaccharides, polyphenols and alcohol epiphytic in Dendrobium officinale in the way of intergrouth under forest was the highest, respectively as 27.29%,0.369% and 7.4%. The amino acids and flavonoids content in Dendrobium officinale in the way of bed planting under forest was the highest, respectively as 4.97% and 1.82%. The experiment result of different cultivation patterns of Dendrobium officinale’s active ingredients showed that the content of polysaccharide and amino acid took the largest proportion in several major components, and the effective composition in the way of the intergrouth under forest and bed planting under forest were the most.2. Based on Dendrobium officinale having antioxidant effect, scavenging free radical effect, compared to Dendrobium officinale extract scavenging ability of free radicals in bed planting under forest, suspension under forest, intergrouth under forest and bed planting in greenhouse, results showed that:(1) the Dendrobium officinale extract by intergrouth under forest on scavenging superoxide anion ability was higher than other three, the Dendrobium officinale extract by canopy bed plant was the worst. (2) The Dendrobium officinale extract by bed planting under forest on scavenging hydroxyl free radical ability was the best, at concentrations for 2~8 mg·mL-1, The Dendrobium officinale extract by bed planting in greenhouse was better than hanging; at a concentration of 8~14 mg·mL-1, the result was opposite. (3) The Dendrobium officinale extract by intergrouth under forest in reducing capacity was higher than other three. (4) The Dendrobium officinale’s DPPH free radical scavenging abilityextract in 3 kinds of imitation wild cultivation didn’t show significant difference,which were nearly 100%.3. By the flat filter paper diffusion method to determined the Dendrobium officinale extract in 4 kinds of cultivation patterns of 5 kinds of tested bacterial inhibition zone diameter, and the double dilution method was adopted to antimicrobial minimum bacteriostatic concentration (MIC) and minimum bactericidal concentration (MBC), the results showed that the Dendrobium officinale extract from different cultivation patterns of 5 kinds of the test bacteria had good inhibitory effect. The antimicrobial activity of Dendrobium officinale extract by intergrouth under forest has the best antimicrobial effect for staphylococcus aureus、the escherichia coli and serratia marcescens H30.When the concentration was 16 mg·mL-1, Antimicrobial effects was 28.59,23.61 and 14.33mm respectively, minimum inhibitory concentration (MIC) was 15.62,31.25 and 250 μg·mL-1. The Dendrobium officinale by bed planting under forest has the best antimicrobial effect for pseudomonas aeruginosa and serratia marcescens MG1.When the concentrateion was 16 mg·mL-1, Antimicrobial effects was 16.67 and 14.97mm respectively,the MIC was 62.5 and 125 μg·mL-1. As we can tell, cultivation patterns affected the bacteriostasis effect of Dendrobium officinale.4. Experimental mice were randomly divided into normal control group, model control group, positive control group and the different cultivation patterns of Dendrobium officinale extract low, medium and high dose group (50、100、200 mg·kg-1). Continuous lavage with 50% ethanol in mice as acute alcoholic liver damage model, the different cultivation patterns of Dendrobium officinale extract continuous lavage for 3 weeks and then sampling, weighing the mice and the liver, detect mice serum index (AST、ALT、ALP、TBIL、TC、TG) and antioxidant index (SOD、MDA、GSH、GSH-Px、ADH、ALDH), and observed the histopathological changes in liver injury of mice. Results showed, Compared with model group, The protective effect of 4 kinds of cultivation patterns of Dendrobium officinale extract on acute liver injury in mice to a certain extent. Among them, the canopy bed plant extracts had the worst activity of acute alcoholic liver injury, the best was intergrouth under forest. The Dendrobium officinale extract of high and middle dose could decrease serum of ALT, AST, ALP activity and the levels of TG and enhance the GSH-PX activity in the liver tissue from 4 kinds of cultivation; The Dendrobium officinale extract of high and middle dose group could reduce TC and MDA content and improve the liver GSH, SOD, ADH and ALDH activity in 3 kinds of wild cultivation. Dendrobium officinale extract can inhibit the acute alcoholic liver injury, this may be related to different growing environment of Dendrobium officinale, which lead to the differences in secondary metabolism.In conclude, The effective component content and pharmacological effects were related to cultivation patterns. Comparison of Dendrobium officinale Kimura et Migo from different cultivation patterns, the effective component content、the vitro antioxidant capacity、the antibacterial effect and the protection on acute alcoholic liver by imitation wild cultivation was superior to the greenhouse cultivation.
